摘 要:为了探究不同冷冻稀释液及抗氧化剂对金华猪精液冷冻效果的影响,试验通过手握法采集健康的20月龄纯种金华猪公猪精液,采用两种常用的商品冷冻稀释液(A、B)稀释精液,然后液氮冻存1 d,解冻后测定精液精子活力、顶体完整率和质膜完整率指标,筛选最适金华猪精液冷冻保存的稀释液;在最适冷冻稀释液中添加20,30,40,50μmol/L表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG),以未添加EGCG和上述添加EGCG的冷冻稀释液稀释金华猪精液,然后液氮冷冻保存精液1 d,解冻后测定精液精子活力、顶体完整率、质膜完整率及抗氧化指标[包括活性氧(reactive oxygen species,ROS)、丙二醛(malondialdehyde,MDA)含量及超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)活性],筛选EGCG的最适添加量。结果表明:两种商品冷冻稀释液的精子活力、顶体完整率、质膜完整率差异不显著(P>0.05),但考虑到价格成本,选择B冷冻稀释液作为后续试验的冷冻稀释液。随着EGCG添加量的增加,金华猪精子的抗氧化能力大致呈现先增加后减少的趋势,其中当EGCG添加量为40μmol/L时,精子活力和顶体完整率显著高于未添加和其他添加量(P<0.05),精子质膜完整率与未添加和其他添加量差异不显著(P>0.05);ROS和MDA含量最低,除MDA含量与30μmol/L差异不显著外(P>0.05),均显著低于未添加和其他添加量(P<0.05);SOD和CAT含量最高,均显著高于未添加和20,30μmol/L,但与50μmol/L差异不显著(P>0.05)。说明在兼顾冷冻效果与成本的基础上,选用加入40μmol/L抗氧化剂EGCG的B冷冻稀释液可以有效改善金华猪冷冻后精液品质,并提高了精子的抗氧化能力。In order to investigate the effects of different freezing dilutions and the addition of antioxidants on semen freezing effects of Jinhua pigs,the semen of healthy 20-month-old purebred Jinhua pig was collected by hand-holding in this experiment.The semen was diluted with two commonly used commercial freezing dilution solutions(A and B).Then,the semen was frozen in the liquid nitrogen for one day.After thawing,the sperm viability,acrosome integrity and plasma membrane integrity were measured to select the most suitable diluent for freezing Jinhua pig semen.The 20,30,40,and 50μmol/L epigallocatechin gallate(EGCG)were added into the optimal freezing diluent.Jinhua pig semen was diluted with EGCG-free and EGCG-added freezing diluents,and then placed in liquid nitrogen for cryopreservation for one day.After thawing,the sperm viability,acrosome integrity,plasma membrane integrity and antioxidant indexes(reactive oxygen species[ROS]and malondialdehyde[MDA]content,superoxide dismutase[SOD]and catalase[CAT]activity)of the semen were measured to select the optimum addition level of EGCG.The results showed that there were no significant differences in sperm viability,acrosome integrity rate and plasma membrane integrity rate between the two commercial cryo-dilutions(P>0.05).However,considering the price and cost,the frozen diluent B was selected as the frozen diluent for the subsequent experiments.With the increase of EGCG addition level,the antioxidant effect of Jinhua pig sperm roughly showed a trend of first increasing and then decreasing.When the EGCG addition level was 40μmol/L,the sperm viability and acrosome intactness were significantly higher than those in the groups without EGCG addition and with other addition levels(P<0.05),but there was no significant difference in the sperm plasma membrane integrity among all groups(P>0.05).The ROS and MDA contents in the 40μmol/L group were the lowest,which were significantly lower than those in other groups(P<0.05)except that MDA content was not significantly different fr
