摘　　要：为了筛选德保猪性成熟前后睾丸中差异表达蛋白(differentially expressed proteins,DEPs),试验选取相同饲养环境下的健康且无亲缘关系的1月龄(性成熟前)与6月龄(性成熟后)德保公猪各3头,采集它们的睾丸并提取蛋白质,利用同位素标记相对和绝对定量(isobaric tags for relative and absolute quantification,iTRAQ)技术对性成熟前后德保猪睾丸中DEPs进行分析,并对DEPs在GO、KOG、KEGG数据库中进行功能注释及通路分析。结果表明:在性成熟前后德保猪睾丸中共鉴定7767个蛋白,筛选出611个DEPs,其中在性成熟后德保猪中显著上调的DEPs 347个,显著下调的DEPs 264个;397个蛋白在GO数据库中得到注释,并富集到53条GO term中,包括细胞过程、生物调节、代谢过程、催化活性等信号通路;409个蛋白在KOG数据库中得到注释,并富集到24种功能,包括信号转导机制、翻译后修饰、蛋白周转、监督等功能;329个蛋白在KEGG数据库得到注释,并富集到287条通路中,包括代谢途径通路、吞噬体、补体与凝血级联反应、肺结核、PI3K-Akt通路等;筛选出4个与繁殖相关的DEPs,包括脂联素(adiponectin)、AQP7、ODF1、磷酸甘油酸激酶2(PGK2),这4个DEPs在精子发生和成熟、保护精子完整性及维持精子活力与功能方面起重要作用。In order to screen the differentially expressed proteins(DEPs),in the testis of Debao pigs before and after sexual maturation,three 1-month-old(before sexual maturation)and three 6-month-old(after sexual maturation)healthy and unrelated Debao pigs were selected in the same feeding environment.The testes were collected and proteins were extracted.The DEPs of Debao pigs before and after sexual maturation were analyzed by iTRAQ(isobaric tags for relative and absolute quantification),and GO,KOG,KEGG databases were used for functional annotation and pathway analysis of the significantly DEPs.The results showed that a total of 7767 proteins were identified in the testis of Debao pigs before and after sexual maturation.A total of 611 DEPs were screened,among which 347 DEPs were significantly up-regulated and 264 DEPs were significantly down-regulated in Debao pigs after sexual maturation;397 DEPs were annotated in the GO database and enriched into 53 GO terms,including cellular processes,biological regulation,metabolic processes and catalytic activity.409 proteins were annotated in the KOG database and enriched to 24 functions,including signal transduction mechanisms,post-translational modifications and protein turnover supervision;329 proteins were annotated in KEGG database,and 287 pathways were enriched,including metabolic pathways,phagosome complement and coagulation cascade,tuberculosis and PI3K-Akt pathway.Four important DEPs related to reproduction were screened,including adiponectin,AQP7,ODF1 and phosphoglycerate kinase 2(PGK2),which played an important role in the spermatogenesis and maturation,protection of sperm integrity,and maintenance of sperm viability and function.
