养肝解郁颗粒对绝经后大鼠非酒精性单纯性脂肪肝脂代谢及肝细胞脂肪变性的影响

摘　　要：目的通过实验研究验证养肝解郁颗粒对绝经后大鼠非酒精性单纯性脂肪肝脂代谢及肝细胞脂肪变性的影响。方法采用大鼠去氧乙烯基环乙烯腹腔注射法来建立绝经期动物模型,通过饲喂高脂高糖饲料制备非酒精性单纯性脂肪肝模型,诱导建立绝经期非酒精性单纯性脂肪肝大鼠动物模型。预防性给药12周后,检测血清AST、ALT、TC、TG水平,计算肝系数,检测肝组织中TC、TG、MDA、SOD、FFA含量,肝脏病理组织学检查。结果养肝解郁颗粒6.0、3.0、1.5 g/kg给药组血清及肝组织匀浆中TC、TG含量、血清AST活性均明显低于模型组(为P<0.01或P<0.05);6.0、3.0 g/kg给药组大鼠血清ALT、肝组织匀浆中MDA、FFA含量、肝脏系数均明显低于模型组(为P<0.01或P<0.05);1.5 g/kg给药组大鼠肝组织匀浆中MDA含量低于模型对照组(为P<0.05);6.0 g/kg给药组大鼠肝组织匀浆中SOD活性明显高于模型对照组(为P<0.05)。养肝解郁颗粒3个剂量均可不同程度减轻肝脏病理损伤。结论养肝解郁颗粒可以调节绝经后大鼠非酒精性单纯性脂肪肝脂代谢、减轻肝细胞脂肪变性程度,从而达到治疗非酒精性单纯性脂肪肝的目的。Objective To verify the effect of Yanggan Jieyu Granules on the lipid metabolism and hepatocyte steatosis in postmenopausal rats with nonalcoholic simple fatty liver with experimental studies.Methods A menopausal animal model was established by intraperitoneal injections of deoxyethylene and cycloethylene in rats,and a model of nonalcoholic simple fatty liver was prepared by feeding rats high-fat and high-sugar diets,and then they were induced to establish a menopausal rat model of nonalcoholic simple fatty liver.After 12 weeks of preventive administration,the levels of serum AST,ALT,TC and TG were detected,the liver coefficient was calculated,and the contents of TC,TG,MDA,SOD and FFA in liver tissues were measured,and a histopathology was performed.Results The contents of TC and TG in serum and liver tissue homogenate and the serum AST activity in the Yanggan Jieyu Granules 6.0g/kg,3.0 g/kg and 1.5 g/kg groups were significantly lower than those in the model group(P<0.01 or P<0.05).The contents of ALT in serum,MDA and FFA in liver tissue homogenate and the liver coefficients in 6.0 g/kg and 3.0 g/kg administration groups were significantly lower than those in the model group(P<0.01 or P<0.05).The MDA content in the 1.5 g/kg administration group was lower than that in the model control group(P<0.05).The SOD activity in liver tissue homogenate of the 6.0 g/kg group was significantly higher than that of the model control group(P<0.05).All the three doses of Yanggan Jieyu Granules reduced the liver pathological injury to different degrees.Conclusion Yanggan Jieyu Granules can regulate the lipid metabolism of nonalcoholic simple fatty liver in postmenopausal rats and reduce the degree of hepatocyte steatosis,thus achieving the goal of treating nonalcoholic simple fatty liver.
