补肾安胎冲剂含药血清对复发性流产小鼠胎盘滋养细胞功能、血管内皮生长因子及其受体表达的影响

摘　　要：目的观察补肾安胎冲剂含药血清对复发性流产(RSA)小鼠胎盘滋养细胞功能、血管内皮生长因子(VEGF)及其受体VEGF-R1、VEGF-R2表达的影响,探讨其治疗RSA的作用机制。方法RSA模型小鼠(CBA/J×DBA/2)和正常妊娠小鼠(CBA/J×BALB/c)分离培养胎盘滋养细胞,将细胞分为正常对照组(正常妊娠小鼠胎盘滋养细胞+空白血清)、模型组(RSA小鼠胎盘滋养细胞+空白血清)和补肾安胎冲剂组(RSA小鼠胎盘滋养细胞+最佳浓度补肾安胎冲剂含药血清),CCK8法检测胎盘滋养细胞增殖活力,流式细胞术检测细胞周期,RT-qPCR检测细胞VEGF、VEGF-R1、VEGF-R2 mRNA表达,Western blot检测细胞VEGF、VEGF-R1、VEGF-R2蛋白表达。结果与正常对照组比较,模型组胎盘滋养细胞增殖活力明显降低(P<0.05),S期细胞比例明显减少,G2/M期细胞比例明显增加(P<0.05),VEGF、VEGF-R1、VEGF-R2mRNA和蛋白表达明显降低(P<0.05);与模型组比较,补肾安胎冲剂组胎盘滋养细胞增殖活力明显增加(P<0.05),S期细胞比例明显增加,G2/M期细胞比例明显减少(P<0.05),VEGF、VEGF-R1、VEGF-R2mRNA和蛋白表达明显升高(P<0.05)。结论补肾安胎冲剂含药血清可增强RSA小鼠胎盘滋养细胞增殖活力,上调VEGF及其受体VEGF-R1、VEGF-R2表达,促进母胎界面血管新生,从而提高妊娠成功率。Objective To observe the effects of Bushen Antai Granules medicated serum placental trophoblast function,VEGF and its receptor expression in recurrent spontaneous abortion(RSA)mice;To explore its mechanism of action in the treatment of RSA.Methods The placental trophoblast cells of RSA model mouse(CBA/J×DBA/2)and normal pregnancy mouse(CBA/J×BALB/c)were isolated and cultured.The cells were divided into normal control group(normal pregnancy mouse placental trophoblast cells+blank serum),model group(RSA mouse placental trophoblast cells+blank serum)and Bushen Antai Granules group(RSA mouse placental trophoblast cells+optimal concentration Bushen Antai Granules containing medicated serum).CCK8 assay was used to detect placental trophoblast proliferation activity,and flow cytometry was used to detect cell cycle,RT-qPCR was used to detect cellular VEGF,VEGF-R1,VEGF-R2 mRNA expressions,Western blot was used to detect VEGF,VEGF-R1 and VEGF-R2 protein expressions.Results Compared with the normal control group,the proliferation activity of placental trophoblast cells in the model group significantly decreased(P<0.05),the proportion of cells in S phase significantly decreased,the proportion of cells in G2/M phase significantly increased(P<0.05),and the mRNA and protein expressions of VEGF,VEGF-R1,and VEGF-R2 significantly decreased(P<0.05).Compared with the model group,the proliferation activity of placental trophoblast cells in the Bushen Antai Granules group significantly increased(P<0.05),the proportion of cells in S phase increased significantly,the proportion of cells in G2/M phase decreased significantly(P<0.05),and the m RNA and protein expressions of VEGF,VEGF-R1,VEGF-R2 significantly increased(P<0.05).Conclusion Bushen Antai Granules medicated serum can enhance the proliferative activity of placental trophoblasts in RSA mice,up regulate the expressions of VEGF and its receptors VEGF-R1 and VEGF-R2,and promote angiogenesis at the maternal fetal interface,so as to improve the success rate of pregnancy.
