摘 要:目的 研究硝菔通结方对慢性传输型便秘(Slow Transit Constipation,STC)的作用机制。方法 用大黄粉混悬液诱导建立SD大鼠STC模型,将大鼠随机分为模型组、硝菔通结方组、莫沙必利组,另设不造模大鼠为空白组。各组大鼠按剂量每日灌胃1次,共28d。空白组、模型组给与同体积纯水干预。苏木素-伊红(Haematoxylin-eosin,HE)染色法观察大鼠结肠组织病理情况;AB-PAS染色观察结肠黏液细胞的黏液分泌情况;蛋白免疫印迹法(Western blot)检测BDNF、TrkB、5-HT、GNDF、GFRα-1、Ret的相关蛋白表达量;实时荧光定量聚合酶链式反应(Real-time PCR)法检测BDNF、TrkB、GNDF、GFRα-1、Ret mRNA表达水平。结果 与空白组比较,模型组大鼠结肠黏膜肿胀,腺体排列杂乱,BDNF、TrkB、5-HT、GNDF、GFRα-1、Ret含量明显降低(P<0.01);与模型组相比硝菔通结方组BDNF、TrkB、5-HT、GDNF、GFRα-1的蛋白表达及mRNA水平均显著升高(P<0.01);Ret mRNA的表达水平也有提升(P<0.05)。结论 硝菔通结方对STC大鼠GDNF与BDNF及其相关蛋白有促进作用。Objective To study the mechanism of action of Nitrogen Tongjie Recipe on chronic transit constipation(Slow Transit Constipation,STC).Methods The SD rat STC model was induced by rhubarb powder suspension,and the rats were randomly divided into model group,Nitrogen Tongjie group and mosapride group,and the rats without model were set as blank group.The rats in each group were intragastrically administered once a day for 28 days.The blank group and model group were given the same volume of pure water intervention.Haematoxylin-eosin(HE) staining was used to observe the histopathological conditions of rat colon;AB-PAS staining was used to observe the mucus secretion of colonic mucous cells;Western blot was used to detect BDNF,TrkB,5-HT,The related protein expressions of GNDF,GFRα-1 and Ret were detected;the mRNA expression levels of BDNF,TrkB,GNDF,GFRα-1 and Ret were detected by real-time quantitative polymerase chain reaction(Real-time PCR).Results Compared with the blank group,the colon mucosa of the model group was swollen,the glands were disorderly arranged,and the contents of BDNF,TrkB,5-HT,GNDF,GFRα-1,and Ret were significantly decreased(P<0.01).Compared with the model group,the protein expressions and mRNA levels of BDNF,TrkB,5-HT,GDNF,and GFRα-1 in the Nitrogen Tongjie Recipe group were significantly increased(P<0.01);the expression level of Ret mRNA was also increased(P<0.05).Conclusion Nitrogen Tongjie Recipe can promote GDNF,BDNF and their related proteins in STC rats.
