摘 要:目的研究独活(Angelicae pubescentis)根的化学成分及其抗骨质疏松活性。方法采用硅胶、RP-18、Sephadex LH-20等层析柱及半制备高效液相色谱对独活根部分95%乙醇提取物进行分离纯化,通过核磁共振波谱、质谱以及与文献数据比较,进行结构鉴定。结果从独活中分离得到20个化合物,分别鉴定为蛇床子素(1),isomeranzine(2),欧芹酚(3),meranzin(4),补骨脂素(5),香柑内酯(6),氧化前胡素水合物(7),(+)-visamminol(8),二氢欧山芹醇乙酸酯(9),二氢欧山芹素(10),二氢欧山芹醇(11),hydroxylomatin(12),当归醇A(13),angelmarin(14),没药当归烯酮(15),decursidate(16),(11S,16S,9Z)-9,17-octadecadiene-12,14-diyne-1,11,16-triol,1-acetate(17),(11S,16S,9Z)-9,17-octadecadiene-12,14-diyne-1,11,16-triol(18),9,12-十八烷二烯酸甲脂(19)和ligustiphenol(20)。结论化合物4,8,12,16,17,18和20为首次从该植物中分离得到。对20个化合物进行抗骨质疏松活性筛选,发现5个化合物(5,6,14,15和18)具有显著抑制RANKL诱导破骨细胞分化的活性。Objective To investigate the chemical constituents of the roots of Angelicae pubescentis and their inhibitory effects on osteoclastic cell formation activity.Methods These compounds were isolated and purified by silica gel,RP-18 column chromatography,Sephadex LH-20 column chromatography and semi-preparative HPLC.Their structures were identified by NMR,MS and comparison with literature data.Results The structure of twenty compounds were identified as osthol(1),isomeranzine(2),osthenol(3),meranzin(4),psoralen(5),bergapten(6),oxypeucedanin hydrate(7),(+)-visamminol(8),columbianetin acetate(9),columbianadin(10),columbianetin(11),hydroxylomatin(12),angelol A(13),angelmarin(14),bisabolangelone(15),decursidate(16),(11S,16S,9Z)-9,17-octadecadiene-12,14-diyne-1,11,16-triol,1-acetate(17),(11S,16S,9Z)-9,17-octadecadiene-12,14-diyne-1,11,16-triol(18),9,12-octadecadienoic acid methyl ester(19),and ligustiphenol(20).Conclusion Compounds 4,8,12,16,17,18 and 20 were firstly isolated from the roots of Angelicae pubescentis.The anti-osteoporosis activity of 20 compounds was screened.Five compounds were found to inhibit RANKL-induced osteoclast differentiation.
