摘  要:目的建立玫瑰花的HPLC指纹图谱和没食子酸、原儿茶酸、原花青素B2、鞣花酸及槲皮素的含量测定方法。方法通过正交试验优选玫瑰花中5种指标成分的提取条件,利用HPLC法建立30批玫瑰花酸水解提取物的指纹图谱,进行相似度评价,并对指认的5个成分进行含量测定,使用SPSS 26.0、SIMCA 14.1等软件对实验数据进行分析评价。结果建立了玫瑰花的指纹图谱,共标定20个共有峰。30批样品的相似度均大于0.9,样品差异不大,质量接近,并筛选出对质量影响较大的成分,分别为峰12(鞣花酸)、峰16、峰19(槲皮素)、峰8、峰3、峰6。5种定量成分在一定范围内线性关系良好(r=0.9994~0.9998),平均回收率(n=6)分别为102.15%、97.59%、98.52%、97.38%、98.62%(RSD<2.7%),平均含量分别为6.63 mg/mL、0.57 mg/mL、7.58 mg/mL、19.33 mg/mL、1.48 mg/mL。结论建立的玫瑰花指纹图谱和5个成分含量测定的方法高效、准确、重复性好,可为玫瑰花药材的质量控制、评价提供参考。Objective To establish the method for HPLC fingerprints of Rosae Rugosae Flos and the contents determination method of gallic acid,protocatechuic acid,procyanidin B2,ellagic acid and quercetin.Methods Orthogonal experiment was used to optimal extraction conditions for 5 quantitative components and the fingerprint of the sample was established by the HPLC.Through similarity evaluation,and SPSS 26.0 and SIMCA 14.1 were used to analyze and evaluate the charateristic chromatogram of 30batches of Rosae Rugosae Flos acid hydrolysate.At last,the content of the 5 identified components was also determined.Results The fingerprint of Rosae Rugosae Flos was established,and 20 common peaks were demarcated.The similarities of the each batches of samples were all above 0.9 comparing with the reference,which indicated little differences between the similarities of samples.And the 6 components of peak 12(ellagic acid),peak 16,peak 19(quercetin),peak 8,peak 3 and peak 6 had a greater impact on the quality evaluation.The 5 quantitative components had a good linear relationship with in a certain range(r=0.9994-0.9998),and the average recovery(n=6)were 102.15%,97.59%,98.52%,97.38%,98.62%(RSD<2.7%),which average content were 6.63 mg/mL,0.57 mg/mL,7.58 mg/mL,19.33 mg/mL,1.48 mg/mL,respectively.Conclusion The established fingerprints of Rosae Rugosae Flos and the content determination method are simple,efficient,accurate and repeatable,which can be used for its quality control.
