摘 要:目的研究不同炮制程度的黄芩炭饮片与浸膏中黄酮类成分的转化规律及有效成分转移率,为黄芩炭的炮制终点判断与质量控制提供参考。方法改变加热温度与时间制备不同炮制程度的黄芩炭饮片,采用煎煮法制备黄芩炭水提浸膏。高效液相色谱法测定黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的含量,分析四种成分的转移率。采用主成分分析对黄芩炭的炮制程度进行综合评价,验证糖苷与苷元的转化模式。结果随着炮制程度的增加,饮片与浸膏中苷元含量均呈现先上升后下降的变化模式,糖苷呈现持续下降的模式,存在糖苷到苷元的转化,同时浸膏中四种成分的转移率逐渐下降。结论炮制程度明显影响黄芩炭中四种黄酮类成分的转化与转移,提示在判断黄芩炭的炮制终点时应关注苷元类含量及成分转移率。Objective To analyze the content transformation of flavonoids and the transfer rate of active components in Scutellariae Radix Charcoal slices and extracts with different processing degrees,and to provide reference for the end point judgment and quality control of Scutellariae Radix Charcoal.Methods Changing the heating temperature and time to prepared different levels of processing scutellariaeradix charcoal slices,using decocting method to prepare Scutellariae Radix Charcoal extracts.The contents of baicalin,wogonoside,baicalein and wogonin were determined by HPLC,and the transfer rate of the four components was analyzed.PCA was used to evaluate the processing degree of Scutellariae Radix Charcoal and verify the transformation mode of glycoside and aglycone.Results With the increase of processing degree,the aglycone content of decoction pieces and extract showed a change rule of first increasing and then decreasing,the glucoside showed a continuous decreasing rule,there was alsoa conversion of glycosides to aglycones,and the transfer rate of four components in extract gradually decreased.Conclusion The degree of processing significantly affect the content transformation and transfer of four flavonoids in scutellaria baicalensis char,suggesting that attention should be paid to the content of aglycones and the rate of component transfer when determining the processing end point of Scutellariae Radix Charcoal.
