河北省猪星状病毒4型全基因组测定及遗传进化分析

摘  要:为了解河北省流行的猪星状病毒(PAstV)遗传特性和重组现象,设计了7对特异性扩增引物,利用RT-PCR方法进行全基因组序列扩增;应用DNAStar软件和MEGA 7.0软件对扩增得到的序列进行拼接和核苷酸同源性分析,并建立基于全基因组和ORF2基因的系统进化树;利用生物学分析软件对全基因组序列结构特点和重组现象进行分析。结果显示,成功扩增得到1株PAstV4 HB-SJZ全基因组序列;全基因组遗传分析显示,HB-SJZ株与PAstV4参考毒株同源性显著高于其他基因型毒株,为74.2%~81.0%,与匈牙利4型毒株WBAstV-1同源性最高(81.0%);基于ORF2基因分析显示,HB-SJZ株与PAstV4参考毒株同源性为64.8%~68.9%,与日本毒株PoAstV-VIRES-GX03-C3遗传关系最近(68.9%);重组分析显示,HB-SJZ株的ORF1基因与JPN Bu52014株和JPN Mol2-32015株存在重组现象。这一研究提示河北省流行的PAstV4是同物种重组毒株,为该病的流行病学调查提供科学依据,也为该病的防控提供理论基础。To understand the genetic characteristics and recombination phenomenon of porcine astrovirus(PAstV)circulating in Hebei Province,seven pairs of specific amplification primers were designed.The whole genome sequence was amplified by RT-PCR.The amplified sequences and nucleotide homology were spliced and analyzed using DNAStar,and phylogenetic trees based on the whole genome and ORF2 gene were established using MEGA7.0 software.The structural characteristics and genetic recombination phenomenon of the whole genome sequence were analyzed by biological analysis software.The results showed that the whole genome sequence of PAstV4 strain was successfully amplified,which possessed the structural characteristics of the astrovirus(AstV)genome.The whole-genome genetic analysis showed that the homology between the HB-SJZ strain and the reference strains of PAstV4 was significantly higher than that of other genotypes,ranging from 74.2%to 81.0%,which shared the highest homology with the type 4 of Hungarian strain WBAstV-1(81.0%).The homology of HB-SJZ strain to PAstV4 was 64.8%-68.9%based on ORF2 gene analysis,and the genetic relationship was the closest with Japanese strain PoAstV-VIRES-GX03-C3(68.9%).Recombination analysis indicated that strain HB-SJZ was the result of novel recombinant events between JPN Bu52014and JPN Mol2-32015,locating at the ORF1gene.The study suggests that the prevalence of PAstV4in Hebei Province is a recombinant strain of the same species,and provides a scientific basis for the epidemiological investigation,and provides a theoretical basis for the prevention and control of the disease.
