摘 要:于2019年4─8月,采用Andersen生物粒子采样器在湖南某地区的生活垃圾中转站连续采集细菌生物气溶胶样品,采用PacBio Sequel测序平台对生活垃圾中转站空气中的细菌群落结构和多样性进行了研究,并分离和纯化了优势菌株,同时对环境因子和微生物群落进行了冗余分析(Redundancy Analysis,RDA)。结果表明:在门水平上,所有样品均主要含有变形杆菌门(Proteobacteria,其物种相对丰度为12.9%~78.0%),其次为厚壁菌门(Firmicutes)和放线菌门(Actinobacteria);在细菌属水平上,有不动杆菌属(Acinetobacter)、芽孢杆菌属(Bacillus)、金黄杆菌属(Chryseobacterium)、假单胞菌属(Pseudomonas)和葡萄球菌属(Staphylococcus)等;初步判定优势菌株为微球菌属的H-1、葡萄球菌属的F-1、埃希氏菌属的F-2和芽孢杆菌属的B-1;空气中存在潜在或机会性致病菌;环境温度和相对湿度对细菌群落结构多样性和细菌群落丰度都有不同程度的影响。Bacterial bioaerosol samples were collected continuously by Andersen biological particle sampler at the refuse transfer station in Hunan Province from April 2019 to August 2019.The community structure and diversity of culturable bacteria in the bioaerosol of the refuse transfer station were studied by Pacbio Sequel sequencing platform,and the dominant strains were isolated and purified.Redundant analysis(RDA)of environmental factors and microbial community was also performed.The results showed that the main microbial groups in the refuse transfer station at the phylum level were Proteobacteria(the species relative abundance of 12.9%~78.0%),followed by Firmicutes and Actinobacteria.The main groups of microorganisms at the genus level were Acinetobacter,Bacillus,Chryseobacterium,Pseudomonas,Staphylococcus,and so on.The dominant bacteria were identified as Micrococcus(H-1),Staphylococcus(F-1),Escherichia(F-2)and Bacillus(B-1).Potential or opportunistic pathogens were found in the air.Air temperature and relative humidity had different effects on the diversity and abundance of bacterial community.
