摘 要:从EGSB反应器的上、中、下三个部位采集污泥样品并持续12个月。宏基因组测序共获得3360条非冗余病毒序列,基于样品内病毒丰度进行群落α和β多样性分析,结果显示:样品内Chao1指数在12个月份间呈现规律性变动趋势;不同月份内样品各自聚类,且来源于春天和冬天的样品病毒群落更相似。噬菌体目(Caudovirales)中的长尾噬菌体科(Siphoviridae)(35.91%±7.84%)、短尾病毒科(Podoviridae)(13.35%±4.54%)和肌病毒科(Myoviridae)(10.44%±2.54%)丰度最高,且体系中存在一群未知病毒科。预测到的主要病毒宿主包括变形杆菌门(Proteobacteria)与放线杆菌门(Actinobacteria)等。7条噬菌体具有调控氮代谢的功能,包含亚硝酸盐还原酶基因(Nitrite reductase,nirK)、羟胺还原酶基因(Hydroxylamine reductase,hcp)或天冬酰胺合成酶(Asparagine synthase,asnB),且整体上十月和十一月拥有氮代谢相关AMG功能的病毒丰度最高。综上,加深了对EGSB体系内病毒群落结构及功能变异模式的理解,扩充了连续运行一整年的WWTP内病毒组动态变化的理论基础。Samples from the upper, middle, and lower layers of the EGSB tank were collected monthly, while 3,360 non-redundant viral sequences were detected. We found that the Chao1 index had a regular changing trend throughout the year and the viral communities were clustered according to the sampling months, while the viral composition in spring and winter seemed more similar. Siphoviridae(with average ± SD value of 35. 91% ± 7.84%), Podoviridae(13. 35%± 4. 54%), and Myoviridae(10. 44% ± 7. 84%) were the most abundant viral families in all samples, which all belonged to the order of Caudovirales. Further, the high diversity and novelty of viral families were observed. The predicted host phyla mainly included Proteobacteria and Actinobacteria. By the annotation of auxiliary metabolic genes(AMGs), seven phages were predicted to be candidates in nitrogen metabolism, owning the nitrite reductase gene(nirK), hydroxylamine reductase(hcp), or asparagine synthase(asnB) in their genome sequences. In particular, the abundances of the viruses containing nitrogen-metabolic AMGs were higher in October and November than that in the other months of the year. Collectively, this study deepens the understanding of the virus community structure and functional variations in the EGSB system, which expands the theoretical basis of the dynamic changes of the virus group in WWTP during continuous operation for a whole year.
