摘 要:目的 建立一种简便、敏感的检测指示系统用于临床样本还原性的分析。方法 以1%浓度碘乙醇-水混合溶液与1%淀粉做梯度正交配比,观察570 nm吸光度变化;以临床药物浓度范围的维生素C及止血敏各与合适碘乙醇-水混合溶液做正交配比,观察570 nm吸光度变化。结果 0.031%~0.036%碘液与1%淀粉(V:V=20:1)混合溶液呈色反应效果最好,且避光条件下较为稳定;浓度为10 mg/L~80 mg/L维生素C与上述浓度碘-淀粉指示系统反应成线性关系;酚磺乙胺浓度越高,对碘-淀粉溶液的消色反应越明显。结论 本文建立的以碘-淀粉溶液为指示剂,检测临床生化样本还原性操作简便,且敏感性与重复性适于临床样本还原性的分析,对于及时发现还原性药物对生化反应的潜在干扰,提高基于Trinders反应的临床生化检验的准确性具有重要的意义。Objective This paper aims to establish a simple and sensitive indicator system for reducibility analysis of clinical samples. Methods The gradient orthogonal ratio of 1% iodine ethanol water mixed solution and 1% starch was used to observe the change of absorbance at 570 nm;vitamin C and hemostatic sensitivity in the concentration range of clinical drugs were mixed with appropriate iodine ethanol water solution to observe the change of absorbance at 570 nm. Results The mixed solution of 0.031%-0.036% iodine solution and 1% starch(V:V=20:1) had the best color reaction effect, and it was relatively stable under the condition of avoiding light. There was a linear relationship between vitamin C concentration of 10 mg/L-80 mg/L and the reaction of iodine starch indicator system. The higher the concentration of etamsylate, the more obvious the achromatic reaction to starch iodine solution. Conclusion The detection system established in this paper, which uses starch iodine solution as an indicator to detect the reducibility of clinical biochemical samples, is simple to operate, and its sensitivity and repeatability are suitable for the analysis of the reducibility of clinical samples. It is of great significance for timely detecting the potential interference of reducing drugs on biochemical reactions and improving the accuracy of clinical biochemical tests based on Trinders reaction.
