摘 要:目的研究正己烷亚急性染毒对小鼠肝细胞形态及肝功能相关酶的影响。方法40只昆明种清洁级小鼠根据不同染毒剂量随机分为低、中、高剂量染毒组和对照组,每组10只。采用灌胃的方式进行正己烷染毒,低、中、高剂量染毒组每只小鼠分别给予浓度为500、1000和2000 mg/kg正己烷0.2 ml/d灌胃;对照组给予食用油0.2 ml/d灌胃,1次/d,连续28 d。染毒结束后,对各组小鼠肝组织进行病理学观察,并进行血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、胆碱酯酶(CHE)以及肝组织ATP酶、脂质过氧化指标的测定。结果与对照组相比,高剂量染毒组小鼠出现明显的肝细胞肿胀、坏死,炎性细胞浸润,细胞间分界模糊等改变。与对照组相比,高、中、低剂量染毒组小鼠血清ALT、AST活力明显升高,高和中剂量染毒组小鼠血清CHE活力明显降低,差异有统计学意义(P<0.05,P<0.01)。与对照组相比,高剂量染毒组小鼠肝组织Na+-K+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活力均明显降低,差异有统计学意义(P<0.05)。与对照组相比,高、中、低剂量染毒组小鼠肝组织T-SOD活力明显降低,差异有统计学意义(P<0.01);高和中剂量染毒组小鼠肝组织谷胱甘肽过氧化物酶(GSH-Px)活力明显降低,丙二醛(MDA)浓度明显升高,差异有统计学意义(P<0.01)。结论正己烷亚急性染毒所致的脂质过氧化增强,可导致肝细胞形态学的异常及肝功能的损伤。Objective To study the effects of n-hexane subacute exposure on liver cell morphology and liver function-related enzymes in mice.Methods Forty mice in exposed groups were given n-hexane exposure by gavage,with 0.2 ml/d of n-hexane at concentrations of 500,1000 and 2000 mg/kg for the low-,the middle-and the high-dose groups respectively.In contrast,the mice in the control group were given 0.2 ml/d of edible oils by gavage.The exposure was carried out once a day for 28 days.After the exposure,the liver tissues of mice in each group were observed pathologically,and serum ALT,AST,CHE,liver ATPase,and lipid peroxidation indexes were measured.Results Compared with those in the control group,the mice in the high-dose group showed obvious changes such as swelling and necrosis of hepatocytes,infiltration of inflammatory cells,and blurred boundary between cells.Compared with those in the control group,the activities of serum ALT and AST in the high-,the middle-and the low-dose groups were increased significantly,while the activities of serum CHE in the high-and the middle-dose groups were decreased significantly,both of which were statistically significant(P<0.05,P<0.01).Compared with those in the control group,the activities of Na+-K+-ATPase and Ca^(2+)-Mg^(2+)-ATPase in the liver tissue of mice in the high-dose group were decreased statistically significantly(P<0.05).Compared with those in the control group,the activity of T-SOD in the liver tissue of mice in the high-,the middle-and the low-dose groups were decreased statistically significantly(P<0.01),while the MDA concentration in the liver tissue of mice in the high-and the middle-dose groups were increased statistically significantly,and their GSH-Px activity were decreased statistically significantly(P<0.01).Conclusion The enhancement of lipid peroxidation caused by n-hexane subacute exposure can lead to abnormal morphology of liver cells and damage of liver function.
