摘 要:为了研究清道夫受体CD36在鱼类抗细菌免疫调控中的作用,本文将鳗弧菌(Vibrio anguillarum)腹腔注射到cd36基因敲除的斑马鱼(cd36^(-/-))和野生型斑马鱼(WT)中,发现突变体斑马鱼的活跃度和存活率均低于野生斑马鱼。分别取鳗弧菌(V.anguillarum)感染的cd36^(-/-)和WT斑马鱼头肾组织进行转录组学测序分析,发现二者差异表达的基因主要富集在补体系统(Complement system)、系统性红斑狼疮(Systemic lupus erythematosus)、细胞因子之间的相互作用(Interaction between cytokines)和病毒致癌机理(Viral carcinogenesis)等与免疫相关的信号途径中。运用实时荧光定量PCR技术进一步证实,与野生型斑马鱼相比,cd36-/-斑马鱼中参与抗原呈递的mhc1和cd8基因、穿孔素基因、补体系统关键基因c3以及细胞因子基因il-4的表达都显著下调。综上,本研究证实CD36在斑马鱼抗鳗弧菌免疫调控中发挥重要作用。CD36 is a single-chain transmembrane glycoprotein,belonging to the class B scavenger receptor family.In fish,the role of CD36 in anti-bacterial immunity remains a subject of investigation.In this study,Vibrio anguillarum was injectedinto the mutant(cd36^(-/-))and the wild zebrafish(WT),and it was found that the mobility and survival rate of the mutants were lower than that of wild zebrafish.Transcriptome analysis of the head-kidney tissues from cd36^(-/-)and wild zebrafish,which were intraperitoneally injected with V.anguillarum,displayed that the genes with significant differences were mainly enriched in the signal pathways of systemic lupus erythematosus,complement system,interaction between cytokines and viral carcinogenesis.Real-time PCR confirmed that,compared to the WT,the antigen presentation-related genes mhc1 and cd8,perforin,complement system key gene c3,and cytokine gene il4 were significantly down-regulated in cd36^(-/-)zebrafish.Collectively,this paper provided evidences for the role of CD36 in regulating the anti-bacterial immunityin fish,and laid a foundation for further study of the immune defense mechanism of CD36 against bacterial infection.
