摘 要:【目的】研究牦牛SMAD4基因SNPs与生长性状的关系,探寻与牦牛生长性状相关的分子标记。【方法】采用DNA测序和单倍型分型技术,对青海牦牛SMAD4基因进行SNPs检测及其基因分型、连锁不平衡和单倍型分析,并对多态位点的基因型及组合单倍型与牦牛生长性状的关联性进行分析。【结果】牦牛SMAD4基因在内含子区域存在6个突变位点,其中g.393A>G、g.555A>G、g.6525A>G和g.18360C>T均存在3种基因型,g.582A>T和g.6492C>T均存在2种基因型。连锁不平衡分析发现,6个位点间不存在强连锁不平衡效应。单倍型分析发现,在14种不同的单倍型中,单倍型H 1的发生频率最高。关联性分析表明,g.393A>G位点与体斜长、胸围和管围均显著相关(P<0.05),g.555A>G位点与体斜长显著相关(P<0.05),g.582A>T位点与体质量和体高均显著相关(P<0.05),g.6492C>T位点与体高显著相关(P<0.05),g.6525A>G位点与体质量显著相关(P<0.05),g.18360C>T位点与胸围显著相关(P<0.05)。基因型组合发现,单倍型H 1H 14可能是影响牦牛生长性状的最优组合。【结论】牦牛SMAD4基因内含子区域上的6个位点与生长性状显著关联,可作为牦牛分子标记辅助选择的候选基因。【Objective】This study analyzed the relationship between SNPs in SMAD4 gene and growth traits to explore the molecular markers related to growth traits in yak.【Method】Using DNA sequencing and haplotyping technology,SNPs,genotyping,linkage disequilibrium and haplotype of SMAD4 gene were analyzed and the relationship between genotype and haplotype combination of polymorphic loci and growth traits of yak was investigated.【Result】There were six mutation sites in the intron region of SMAD4,among which g.393A>G,g.555A>G,g.6525A>G and g.18360C>T had three genotypes,while g.582A>T and g.6492C>T had two genotypes.Linkage disequilibrium analysis showed that there was no strong linkage disequilibrium effect among the 6 loci.Haplotype analysis showed that H 1 had the highest frequency of occurrence among 14 different haplotypes.Correlation analysis showed that g.393A>G locus was significantly correlated with body oblique length,chest circumference and cannon circumference(P<0.05),g.555A>G locus was significantly correlated with body oblique length(P<0.05),g.582A>T locus was significantly correlated with body mass and withers height(P<0.05),g.6492C>T locus was significantly correlated with withers height(P<0.05),g.6525A>G locus was significantly correlated with body mass(P<0.05),and g.18360C>T locus was significantly correlated with chest circumference(P<0.05).The genotype combination found that haplotype H 1H 14 may be the best combination affecting growth traits of yak.【Conclusion】Six loci in the intron region of SMAD4 gene were significantly associated with growth traits and could be used as candidate genes for marker-assisted selection in yaks.
