摘  要:[目的]双苯[口恶]唑酸是一种除草剂安全剂,可缓解禾草灵对水稻和玉米的药害作用,扩大其在水田的使用,应评估双苯[口恶]唑酸对水生生物的毒性。[方法]研究双苯[口恶]唑酸对斑马鱼胚胎的急性毒性、生长发育的影响及影响生长发育的机理。[结果]双苯[口恶]唑酸对斑马鱼的LC_(50)值在48、96 h,分别为1.81、1.66 mg/L。斑马鱼胚胎暴露于0.01、0.1 mg/L的双苯[口恶]唑酸,胚胎孵化时间延长,幼鱼体内SOD(超氧化物歧化酶)、CAT(过氧化氢酶)、MDA(丙二醛)、ALDH(乙醛脱氢酶)、GSH(还原型谷胱甘肽)活性增加,gstp2、cat、bcl2表达量增加,Cu/Zn-sod、Mn-sod和bax表达量减少。[结论]双苯[口恶]唑酸对斑马鱼胚胎的毒性为中毒,胚胎可以通过体内相关基因表达量的变化改变氧化应激酶活性进行解毒。[Aims] Isoxadifen-ethyl is a herbicide safener, which is used to alleviate the harm of diclofop-methyl on rice and maize. Assessing the toxicity of isoxadifen-ethyl in aquatic organism is fundamental to expand its use in paddy fields. [Methods] Acute toxicity of isoxadifen-ethyl on zebrafish embryos and its effects on embryos growth and development were determined, and the mechanism affecting their growth and development was discussed. [Results] The results showed that the LC_(50)(48, 96 h) of isoxadifen-ethyl on zebrafish were 1.81 and 1.66 mg/L. When zebrafish embryos were exposed to 0.01 and 0.1 mg/L isoxadifen-ethyl, the hatched time was prolonged, and there was no significant change in 96 h hatching rate, body length and the number of heartbeats within 60 s. The changes of enzyme and gene expression in young fish were determined to study its mechanism. The results showed that the activities of SOD(superoxide dismutase), CAT(catalase), MDA(malondialdehyde), ALDH(acetaldehyde dehydrogenase) and GSH(reduced glutathione) increased, and so does the expression of gstp2, cat and bcl2, while the expression of Cu/Zn-sod,Mn-sod and bax decreased. [Conclusions] The toxicity of isoxadifen-ethyl to zebrafish embryos is moderate toxicity,and embryos can detoxify isoxadifen-ethyl by changing the activity of oxidative stress enzymes through changes in the expression of related genes.
