摘 要:【目的】探究雌激素受体基因(Estrogen receptor,ESR)在家兔群体中不同性腺组织中的表达规律及其在分子调控机制中所发挥的作用,为利用分子标记技术改良家兔繁殖性状提供科学依据。【方法】对不同繁殖时期的齐卡巨型白兔、加利福尼亚兔和齐兴肉兔的卵巢、输卵管和子宫组织的ESR基因进行实时荧光定量(Real-time PCR)检测。【结果】ESR1基因在的3个不同品种妊娠母兔的卵巢、输卵管和子宫组织中均有不同程度的表达,不同品种和不同组织间差异均不显著。齐卡巨型白兔在不同繁殖时期,3个性腺组织中均有ESR1表达,且表达趋势趋于一致,从高到低为子宫、输卵管、卵巢,进一步对各组织表达量分析发现,ESR1基因在空怀母兔子宫的表达量显著高于妊娠母兔(P<0.05),空怀母兔输卵管的表达量显著高于妊娠母兔(P<0.05)。【结论】在怀孕期齐卡巨型白兔、加尼福尼亚兔和齐兴肉兔的卵巢、输卵管和子宫等组织中均检测到ESR1基因的表达,表明ESR1基因在家兔繁殖过程中可能起重要作用。ESR1基因在齐卡巨型母兔的性腺组织中不同生理阶段的表达量存在差异,可能与该品种的繁殖力有关。【Objective】The study aimed to explore the expression rule of ESR gene in different gonad tissues of domestic rabbit population, as well as its role in molecular regulation mechanism, so as to provide scientific basis for improving reproductive traits of rabbits by using molecular marker technology. 【Method】Against the background, real-time PCR was applied to detect ESR gene in ovary, fallopian tube and uterus tissues of the following rabbits at different reproduction periods, including Zika giant white rabbits, California rabbits and Qixing meat rabbits.【Result】ESR1 gene was expressed in ovaries, fallopian tubes and uterus tissues of pregnant female rabbits from three different breeds to different degrees, while no significant difference existed between different breeds and different tissues. ESR gene was expressed in the three gonads of Zika giant white rabbits at different reproduction periods, and the expression trend tended to be consistent, namely uterus>fallopian tube>ovary. Further analysis on the ESR expression levels in each tissue was carried out, and it was found that the expression of ESR1 gene in uterus of empty pregnant rabbits was significantly higher than that of pregnant rabbits(P < 0.05). The expression of ESR1 gene in empty pregnant rabbits was significantly higher than that of pregnant rabbits(P < 0.05). The expression of ESR1 gene in the fallopian tube of nonpregnant rabbits was significantly higher than that in pregnant rabbits(P < 0.05). 【Conclusion】The expression of ESR1 gene was detected in the ovary, fallopian tube and uterus of Zika giant white rabbits, California rabbits and Qixing meat rabbits during pregnancy, indicating that ESR1 gene may play an important role in rabbit reproduction.There was significant difference in the expression level of ESR1 gene in the gonad tissue of Zika giant female rabbit with different physiological stages. It may be relevant to the reproductive capacity of the breed.
