摘 要:目的研究液氮罐失真空对精子、卵子、胚胎冷冻保存效果的影响。方法在部分失真空液氮罐(A)、完全失真空液氮罐(B)、完好液氮罐(C)加满液氮后,每隔1 h测量液氮挥发量。将小鼠的精子、卵母细胞、卵裂期胚胎经玻璃化冷冻后贮存于B罐中,实时记录液氮罐内温度变化,在温度上升到-180℃时解冻第一组小鼠精子、卵母细胞以及卵裂期胚胎,之后每隔1h解冻一组,分别检测其存活率。结果B罐液氮挥发最快,其次是A罐,最慢是C罐。在B罐保存的精子复苏后24 h内存活率差异无统计学意义(P>0.05);在B罐保存的卵母细胞和卵裂期胚胎的复苏后存活率第4组显著低于前3组(P<0.05)。结论液氮罐失真空后26 h内温度迅速上升,对精子损伤影响较小。但温度上升至-63℃时,对卵母细胞与胚胎会造成不可逆的损伤。Objective To study the effect of vacuum loss of liquid nitrogen tank on cryopreservation of sperm,ovum and embryo.Methods After partial(A),complete(B)and intact(C)tanks were flled with liquid nitrogen,the amount of liquid nitrogen volatilization was measured every 1 h.Mouse sperm,oocytes and embryos at cleavage stage were stored in tank B after vitrification and freezing.Temperature changes in liquid nitrogen tank were recorded in real time.The first group of mouse sperm,oocytes and embryos at cleavage stage were thawed when the temperature rose to-180C,and then one group was thawed every 1 h to detect their survival rates respectively.Results The volatilization rate of liquid nitrogen in tank B was the fasest,fllowed by tank A,and tank C was the slowest.There was no significant difference in the survival rate of sperm stored in B tank within 24 h after resuscitation(P>0.05).The survival rate after resuscitation of oocytes and cleavage stage embryos in B tank in the fourth group was significantly lower than that in the first three groups(P<0.05).Conclusion The temperature of liquid nitrogen tank rises rapidly within 26 h after vacuum loss,which has lttle effect on sperm damage.But when the temperature rises to-63C,it will cause irreversible damage to oocytes and embryos.
