摘　　要：目的探讨circRNA 0000809差异表达对HTR-8/Svneo细胞生长、迁移和侵袭的影响。方法将HTR-8/Svneo培养细胞系分为过表达circRNA 0000809组、空载表达组、空白对照组3组。实时荧光定量PCR法检测circRNA 0000809表达水平,MTT法检测细胞存活率,划线实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,蛋白质印记分析检测E-黏钙蛋白(E-cadherin)和发状分裂相关增强子1(HES-1)表达水平。结果与对照组或空载组比较,过表达组细胞circRNA 0000809表达水平显著升高(P<0.05);circRNA 0000809过表达的细胞增殖水平显著低于空载组及对照组,划线痕迹愈合速度及细胞侵袭能力显著低于空载组及对照组(P<0.05);circRNA 0000809过表达促进细胞中E-cadherin表达,抑制了HES-1的表达(P<0.05)。结论circRNA 0000809过表达可抑制HES-1表达,同时显著抑制滋养层细胞的侵袭和迁移能力,以及上皮-间充质的转化过程,抑制细胞增殖并促进细胞死亡。Objective To investigate the effects of circRNA 0000809 differential expressions on the growth,migration and invasion of HTR-8/Svneo cells.Methods The HTR-8/Svneo cell lines were divided into three groups circRNA 0000809 overexpression group,no-load expression group and control group.The expression level of circRNA 0000809 was detected by real-time fluorescence quantitative PCR,the cell survival rate was detected by MTT,the cell migration ability was detected by streaky assay,the cell invasion ability was detected by Transwell assay,and the expression levels of E-cadherin and HES-1 were detected by protein imprinting analysis.Results Compared with the control group or no-load group,the expression level of circRNA 0000809 in overexpression group was significantly increased,and the difference was statistically significant(P<0.05).The circRNA 0000809 overexpressed cells proliferation level was significantly lower than that in no-load group and control group,line marks healing speed and cell invasion ability were significantly lower than that in no-load group and control group,the difference was statistically significant(P<0.05).The circRNA 0000809 overexpression promoted the expression of E-cadherin and inhibited the expression of HES-1 in cells(P<0.05).Conclusions The circRNA 0000809 overexpression can inhibit HES-1 expression,significantly inhibit the invasion and migration of trophoblast cells,the epithel-mesenchymal transformation process,and cell proliferation and promote cell death.
