摘 要:通过构建昆明裂腹鱼全长转录组文库获得基因全长,并通过设计引物PCR扩增,验证高通量测序的准确性,同时对CYP1A1基因的CDS序列进行基因结构和生物信息学分析。结果表明:昆明裂腹鱼CYP1A1基因的CDS序列为1498 bp,编码299个氨基酸,与鲤鱼、安水金线鲃、鲫鱼的CDS序列的同源性相似比率分别为91.70%、92.23%、91.63%,氨基酸序列同源性相似比率分别为90.55%、90.16%、89.76%,昆明裂腹鱼CYP1A1基因的氨基酸的相对分子质量为55867.48 KD,理论等电点(pI)为6.44,分子式为C_(2505)H_(3965)N_(671)O_(728)S_(23);分子系统进化树分析表明,与金线鲃、犀角金线鲃和安水金线鲃亲缘关系相近,与斑马鱼亲缘关系较远;昆明裂腹鱼CYP1A1基因存在明显的疏水性区域,有1个信号肽、27个磷酸化位点、2个跨膜区、有1个N-糖基化位点,3个O-糖基化位点;二级结构主要以α-螺旋为主,无规则卷曲次之,β-折叠最少;三级结构分析表明,昆明裂腹鱼CYP1A1基因有1个缺失突变位点,6个Cys位点。该研究为进一步研究昆明裂腹鱼CYP1A1基因的功能奠定了基础。The full length of the gene was obtained by constructing the full length transcriptome library of the Schizothorax grahami,and the accuracy of high-throughput sequencing was verified by designing primer PCR amplification.At the same time,the gene structure and bioinformatics analysis of the CDS sequence of the CYP1A1 gene were carried out.The results showed that the CDS sequence of the CYP1A1 gene of the Schizothorax grahami was 1498 bp,encoding 299 amino acids,and the homology similarity ratio of the CDS sequence of Cyprinus carpio,Sinocyclocheilus anshuiensis and Carassius auratus was 91.70%,92.23%,and 91.63%,respectively.The homology similarity ratio of the amino acid sequence was 90.55%,90.16%,and 89.76%,respectively.The relative molecular weight of the amino acid of the CYP1A1 gene of Schizothorax grahami was 55867.48 KD,and the theoretical isoelectric point(pI)was 6.44,The molecular formula is C_(2505)H_(3965)N_(671)O_(728)S_(23);molecular phylogenetic tree analysis showed that it was close to Sinocyclocheilus grahami,Sinocyclocheilus rhinocerous and Sinocyclocheilus anshuiensis,but far from Danio rerio;The CYP1A1 gene of Schizothorax grahami has obvious hydrophobic regions,including 1 signal peptide,27 phosphorylation sites,2 transmembrane regions,1N-glycosylation site and 3 O-glycosylation sites;the secondary structure is mainly composed of α-Spiral is the main,followed by irregular curl,β-folding is the least;the tertiary structure analysis showed that there were 1 deletion mutation site and 6 Cys sites in the CYP1A1 gene of Schizothorax grahami.This study laid a foundation for further study on the function of CYP1A1 gene of Schizothorax grahami.
