温度和盐度对珍珠龙胆石斑鱼血清免疫因子活性的联合效应

摘  要:【目的】研究环境因子温度、盐度同时变化对珍珠龙胆石斑鱼(Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂)血清免疫因子活性的影响,探讨机体抵御温盐变化的生理生化反应规律。【方法】在温度14~36℃、盐度5~45条件下,每个因子各取3个编码水平,设计中心复合实验,构建温度、盐度对珍珠龙胆石斑鱼幼鱼血清过氧化物歧化酶(SOD)活性、溶菌酶(LZM)活性、补体(C3、C4)含量影响的二次回归模型,分析温度、盐度对上述指标的一次、二次效应,采用响应曲面分析法分析温度、盐度对上述指标的联合效应。【结果】构建的温度与盐度对血清免疫因子影响的二次回归模型方程拟合度高;温度、盐度的一次效应、二次效应对于珍珠龙胆石斑鱼幼鱼血清LZM活性、补体C3含量、补体C4含量的影响极为显著(P<0.01),温度的一次效应对于血清SOD活性的影响极为显著(P<0.01);在实验设定的温度、盐度范围内,温度、盐度对血清LZM活性、补体C4含量存在显著的交互效应(P<0.05)。【结论】温度、盐度可显著影响血清免疫因子的活性,并对LZM活性、补体C4含量存在显著的协同效应,表明温度、盐度是影响珍珠龙胆石斑鱼免疫的重要环境因子。【Objective】To study the effects of temperature and salinity on the activity of serum immune factors of pearl gentian grouper(Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂),and to explore the physiological and biochemical reaction rules of the body against changes of temperature and salt.【Method】Under the conditions of temperature(14-36℃)and salinity(5-45),three coding levels were selected for each factor.Through the central composite test design,quadratic regression models were established to analyze the linear and quadratic effects of temperature and salinity on the activities of serum superoxide dismutase(SOD),lysozyme(LZM)and the contents of complement C3/C4 in the young pearl gentian grouper.The response surface analysis method was used to study the combined effects of temperature and salinity on the above indicators.【Results】The quadratic regression model of the effects of temperature and salinity on serum immune factors has a high degree of fit;The linear and quadratic effects of temperature,salinity on LZM complement C3 and complement C4 were highly significant(P<0.01),the linear effects of temperature on SOD enzyme activity was highly significant(P<0.01);Within the range of temperature and salinity set in the test,the interactive effects of temperature and salinity on LZM,complement C4 were significant(P>0.05).【Conclusion】Temperature and salinity can significantly affect the activity of serum immune indicators,and have significant combined effects on the activity of LZM and the content of complement C4,indicating that temperature and salinity were important environmental indicators affecting the immunity of Pearl gentian grouper.
