摘  要:目的利用基于线粒体细胞色素C氧化酶Ⅰ(COⅠ)基因、线粒体控制区(D-loop)基因、16S rRNA基因的DNA条形码技术对广州口岸截获的1只死鼠进行种类鉴定。方法从鼠肌肉中提取DNA,用PCR法扩增COⅠ、D-loop、16S rRNA基因片段,将测序结果与GenBank和BOLD systems v4中鼠类的DNA条形码进行比对,用Mega 6.0构建系统发育树,计算遗传距离。结果该鼠类样本能扩增出COⅠ、D-loop、16S rRNA基因片段,通过COⅠ、16S rRNA基因扩增片段序列比对与德氏柔毛鼠Praomys daltoni同源性均为99%,D-loop基因扩增片段序列比对与猪吻柔毛鼠Praomys rostratus同源性为100%,结合形态学特征,最终鉴定为德氏柔毛鼠Praomys daltoni。结论COⅠ、16S rRNA基因片段能够对鼠类进行有效鉴定,弥补了形态学鉴定的不足之处。Objective To identify one intercepted died rodent by using DNA barcode based on cytochrome C oxidase subunitⅠ(COⅠ)gene,mitochondrial control region(D-loop)gene,16S rRNA gene sequences at Guangzhou port.Methods DNA was extracted from the intercepted rodent muscles.The COⅠ,D-loop,16S rRNA gene fragment were amplified and sequenced.The sequences were compared on GenBank and BOLD systems v4.Phylogenetic tree was constructed and genetic distance was calculated with Mega 6.0 based on analysis of COⅠ,16S rRNA gene data.Results The specific COⅠ,D-loop,16S rRNA gene fragments were successfully amplified by PCR.The comparisons on GenBank and BOLD systems v4 showed that COⅠ,16S rRNA sequences had 99%homology with Praomys daltoni,D-loop Sequence had 100%homology with Promys rostratus.Combined with morphological features,it belonged to Praomys daltoni.Conclusion COⅠ,16S rRNA gene sequences can be used as an effective technique to identify intercepted rodents,it is a powerful complement to morphological identification.
