摘  要:目的探究水合橙皮内酯对阿尔兹海默症大鼠的保护作用及可能机制。方法SD大鼠随机分为假手术组(Sham组)、阿尔兹海默症组(AD组)和水合橙皮内酯组(MH组),每组10只。水迷宫实验检测大鼠空间学习记忆能力;HE染色观察大鼠海马组织病理学变化;尼氏染色观察大鼠海马组织神经元存活;ELISA方法检测大鼠海马组织神经递质及相关因子水平;TUNEL实验检测大鼠海马组织细胞凋亡;Western blot检测大鼠海马组织BDNF、TrkB和p-CREB蛋白表达水平。结果水合橙皮内酯降低阿尔兹海默症大鼠逃避潜伏期,增加大鼠穿越平台次数和海马组织神经元细胞数,降低大鼠海马组织TUNEL阳性细胞数,增加大鼠海马组织BDNF、TrkB和p-CREB蛋白表达水平。结论水合橙皮内酯通过激活BDNF/TrkB/CREB信号通路改善AD大鼠学习记忆能力。Objective To explore the protective effect of meranzin hydrate on Alzheimer’s disease rats and its possible mechanism.Methods SD rats were randomly divided into sham operation group(Sham group),Alzheimer’s disease group(AD group)and meranzin hydrate group(MH group),with 10 rats in each group.Water maze test was used to test the spatial learning and memory ability of rats.The histopathological changes of hippocampus were observed by HE staining.The neuronal survival of rat hippocampus was observed by Nissen staining.The levels of neurotransmitters and related factors in rat hippocampus were detected by ELISA.The cells apoptosis of hippocampal tissue was detected by TUNEL assay.The expression of BDNF,TrkB and p-CREB in hippocampus were detected by Western blot.Results Meranzin hydrate decreased the escape latency of Alzheimer’s disease rats,increased the times of crossing the platform and the number of neurons in the hippocampus,decreased the number of TUNEL positive cells in the hippocampus,and increased the expression of BDNF,TrkB and p-CREB in the hippocampus.Conclusion Meranzin hydrate can improve the learning and memory ability of AD rats by activating BDNF/TrkB/CREB signaling pathway.
