摘 要:目的:观察柴当薄组(CDB)各极性溶剂提取部位对行为绝望模型小鼠的影响,获得其抗抑郁效应良好的提取部位,并开展该部位小鼠急性毒性试验以初步确定其安全性。方法:采用极性梯度萃取法分别获得CDB水煎液与醇提物的石油醚部位、乙酸乙酯部位及正丁醇部位,合计6个CDB提取部位,以21.9g·kg^(-1)连续灌胃给药14d,采用小鼠悬尾试验(TST)和小鼠强迫游泳试验(FST)观察6个CDB提取部位对小鼠不动时间的影响,筛选出CDB抗抑郁有效部位。其次,开展CDB醇提石油醚部位的小鼠半数致死量(LD_(50))及醇提乙酸乙酯部位小鼠最大耐受量(MTD)测定。结果:21.9g·kg^(-1)的CDB醇提石油醚部位及醇提乙酸乙酯部位均能显著缩短TST及FST中的不动时间,且不影响小鼠自主活动;而CDB水煎液的3个极性提取部位及醇提物的正丁醇部位作用不明显。CDB醇提石油醚部位小鼠灌胃给药的LD_(50)为851.4 g·kg^(-1),约临床日用剂量的1554倍;醇提乙酸乙酯部位小鼠灌胃的MTD为394.2g·kg^(-1),约临床日用剂量的720倍。结论:CDB醇提石油醚部位和醇提乙酸乙酯部位是其发挥抗抑郁作用的主要有效部位,且两提取部位安全性较高。Objective:To observe the effects of each polar solvent extraction fraction of ChaiDangBo group(CDB)on behavioral desperation model mice,and obtain its extraction fraction with good antidepressant effects,as well as to conduct acute toxicity tests in mice for the fraction to initially determine its safety.Method:The polar gradient extraction method was used to obtain the petroleum ether,ethyl acetate and n-butanol fractions of CDB water decoction and alcoholic extract,respectively,amount to 6 CDB extract fractions,which were given to mice by gavage for 14 d with a dose of 21.9 g·kg^(-1).The effects of the 6 CDB extract fractions on the immobility time of mice were observed by tail suspension test(TST)and forced swimming test(FST),and the effective fractions for antidepressant of CDB were screened.Furthermore,the determination of mouse LD_(50) of the ethanolextracted petroleum ether fraction of CDB and mouse maximum tolerance dose(MTD)of the ethanol-extracted ethyl acetate fraction were carried out.Result:21.9 g·kg^(-1)of ethanol-extracted petroleum ether fraction and ethanol-extracted ethyl acetate fraction of CDB both significantly shortened the immobility time in the TST and FST,and did not affect the voluntary activities of mice.While the three fractions from CDB water decoction and the ethanol-extracted n-butanol fraction had no significant effects.The LDso of ethanol-extracted petroleum ether fraction administration by gavage for mice was 851.4 g·kg^(-1),about 1554 times of the clinical daily dose.The MTD of ethanol-extracted ethyl acetate fraction for mice was 394.2g·kg^(-1),about 720 times of the clinical daily dose.Conclusion:The ethanol-extracted petroleum ether fraction and ethanol-extracted ethyl acetate fraction are the main effective fractions of CDB for its antidepressant effect,and the safety of the two extract fractions is high.
