摘  要:肌醇半乳糖苷合酶(galactinol synthase, GolS)是棉子糖家族寡糖(raffinose family oligosaccharides, RFOs)生物合成中的关键酶,在植物响应非生物胁迫的过程中发挥重要作用。本研究通过生物信息学的方法,基于本团队组装的耐寒优质苹果(Malus pumila)品种‘寒富’的基因组,对苹果Gol S基因家族成员进行鉴定、比较与表达分析。结果表明,‘寒富’基因组中共有42个MdGolS成员,它们分布于16条染色体上。系统发育分析表明, MdGolS基因家族成员分为三个亚家族,相同亚家族成员的保守基序的分布和种类较为一致。共线性分析表明,大片段复制现象是MdGolS基因家族扩增的主要动力。分析发现GolS基因启动子序列含有多种与植物生长发育、胁迫反应等相关的应激反应元件和激素响应元件。此外,结合冷冻处理的转录组数据发现,与不耐寒性品种‘长富2号’相比, MdGolS15、MdGolS22、MdGolS27、MdGolS35和MdGolS42仅在耐寒性品种‘寒富’中受到低温胁迫的强烈诱导,推测可能在响应低温中发挥重要作用。GolS基因家族的鉴定与分析为研究低温诱导RFOs合成代谢增强苹果耐寒性的机制提供研究基础。Galactinol synthase(GolS) is a key enzyme in raffinose family oligosaccharides(RFOs) biosynthesis, which plays an important role in plant response to abiotic stress. In this study, the Gol S gene family members of apple were identified, compared and analyzed based on the genome of cold-tolerant and high-quality apple(Malus pumila) cultivar ’Hanfu’ assembled by our research group through bioinformatics methods. The results showed that there were 42 Md Gol S members in the ’Hanfu’ genome, which were distributed on 16 chromosomes. Phylogenetic analysis showed that Md Gol S were divided into three subfamilies, and the distribution and type of conserved motifs of the same subfamily members were relatively consistent. Synteny analysis showed that large fragment replication was the main driving force for the amplification of Md Gol S gene family. Analysis of the Gol S gene promoter sequence revealed that it contains a variety of stress response elements and hormone response elements related to plant growth and development or stress response. In addition, combined with the RNA-seq data of freezing treatment, it was found that compared with the cold-intolerant cultivar ’Naganofuji No. 2’, MdGolS15, MdGolS22, MdGolS27,MdGolS35 and MdGolS42, were strongly induced by low temperature stress only in the cold-tolerant cultivar ’Hanfu’, which may play an important role in responding to low-temperature. The identification and analysis of Gol S gene family provide a basis for the study of the mechanism of cold tolerance of apple enhanced by RFOs synthesis induced by low temperature.
