摘 要:本文对芍药(Paeonia lactiflora)花药培养及植株再生影响因素进行研究,以期为芍药遗传理论研究及实际育种提供新材料和技术支持。以4个芍药品种为材料,研究了低温预处理时间、黑暗培养时间、2,4-D浓度对愈伤组织诱导的影响,并比较了4个品种愈伤组织诱导率、体胚发生率和成苗率差异。结果表明:低温预处理6~8 d及黑暗培养0~7 d能提高愈伤组织诱导率,不同品种对2,4-D浓度需求不同,‘鱼鳞红’和‘东海朝阳’以0.5 mg·L^(-1)为宜,‘大富贵’和‘粉玉奴’以1.5 mg·L^(-1)为宜;仅‘大富贵’和‘粉玉奴’获得再生植株,包括单倍体、二倍体、非整倍体植株,以及少量的三倍体和四倍体植株等多种类型。因此,选择合适低温预处理时间和黑暗培养时间有利于提高花药愈伤组织诱导率,不同品种对芍药花药离体培养效果不同。The influencing factors of haploid formation induced by anther of peony(Paeonia lactiflora) were studied to provide new materials and technical support for the theoretical study on plant genetics and practical breeding. Taking the anther from four peony varieties as explants, the effects of low-temperature pretreatment period, dark culture period, 2,4-D concentration on callus induction were studied, and the callus induction rate, somatic embryo rate, seedling rate of the four peony varieties were compared. The results showed that low-temperature pretreatment for 6—8 d and dark culture for 0—7 d had a positive effect on callus induction. The amount of 2,4-D added in the medium was different with different peony varieties,0.5 mg·L^(-1)was appropriate for ’Yulinhong’ and ’Donghaichaoyang’, 1.5 mg·L^(-1)was appropriate for ’Dafugui’and ’Fenyunu’. Only ’Dafugui’ and ’Fenyunu’ could obtain regenerated plants, including haploid, diploid, aneuploid plants, and a small number of triploid and tetraploid plants. Therefore, choosing appropriate low-temperature pretreatment period and dark culture period is beneficial to increase the induction rate of anther callus, and different varieties have different effects on the in vitro culture of peony anthers.
