摘  要:目的:探讨脂肪酸转位酶(fatty acid translocase,FAT/CD36)对人肺动脉内皮细胞(human pulmonary artery endothelial cells,HPAEC)的增殖及凋亡的影响。方法:(1)收集20例肺动脉高压(pulmonary arterial hypertension,PAH)患者及20例正常人血清,用酶联免疫吸附试验检测并比较两组血清中CD36的浓度。(2)慢病毒转染HPAEC,并通过Western Blot和实时荧光定量聚合酶链式反应验证转染效率,细胞计数实验检测细胞增殖,流式细胞术检测细胞凋亡。结果:(1)PAH患者血清中CD36平均浓度显著低于正常组(P<0.05)。(2)在HPAEC中分别转染过表达、敲减病毒,成功构建稳定转染的细胞株。(3)与对照组相比,CD36过表达后,细胞凋亡率显著增加,细胞增殖速度显著降低,而敲减CD36显著抑制了细胞凋亡。结论:CD36通过参与HPAEC增殖及凋亡,从而影响PAH的发生发展。Objective:To investigate the effect of fatty acid translocation enzyme(FAT/CD36)on pulmonary artery endothelial cells(HPAEC)and its possible related mechanism.Methods:(1)Serum levels of CD36 in 20 pulmonary arterial hypertension(PAH)patients and 20 normal controls were detected by enzyme linked immunosorbent assay and compared.(2)Lentivirus transfected HPAEC,and the transfection efficiency was verified by Western Blot and real-time quantitative polymerase chain reaction.Cell proliferation was detected by cell counting kit-8 assay,and cell apoptosis was detected by flow cytometry.Results:(1)The mean concentration of CD36 in serum of PAH patients was significantly lower than that of normal group,the difference was statistically significant.(2)Overexpressed and knockdown viruses were transfected in HPAEC respectively,and stably transfected cell lines were successfully constructed.(3)Compared with the negative control group,CD36 overexpression significantly increased cell apoptosis rate and decreased cell proliferation rate,while knockdown CD36 significantly inhibited cell apoptosis.Conclusion:CD36 affects the development of PAH by participating in HPAEC proliferation and apoptosis.
