摘 要:研究了白玉蜗牛黏液中糖胺聚糖的化学结构及其对血液凝固的影响.采用酶解法提取白玉蜗牛黏液粗多糖,季铵盐沉淀法和离子交换柱层析法纯化获得多糖SMG-2.单糖组成分析以及紫外、红外、核磁共振等波谱方法对SMG-2的化学组成与结构进行分析研究,体外抗凝血实验分析SMG-2对人质控血浆凝血时间的影响.结果表明,SMG-2由L-艾杜糖醛酸和D-乙酰氨基葡萄糖组成的类肝素糖胺聚糖,因其肝素抗凝五糖序列而无显著抗凝血活性.The chemical structure of glycosaminoglycan from the snail Achatina fulica mucus and its anticoagulation were investigated.The crude polysaccharide SMG-1 were extracted from the snail mucus by enzymatic hydrolysis,and the purified polysaccharide SMG-2 was obtained by quaternary ammonium salt precipitation and ion exchange column chromatography.The chemical structure of SMG-2 was clarified by monosaccharide composition analysis and UV,IR and NMR spectroscopy.The anticoagulant activity of SMG-2 was evaluated by in vitro using human quality control plasma.The results showed that SMG-2 was a heparin-like glycosaminoglycan which was composed of iduronic acid and N-acetyl-glucosamine,and had no significant anticoagulant activity due to lacking of heparin-pentosaccharide sequence.
