摘 要:体内NAD^(+)会随着机体衰老出现系统性减少,从而引发年龄有关的生理功能下降和疾病发生.补充NAD^(+)的中间体β-烟酰胺单核苷酸(β-nicotinamide mononeucleotide,β-NMN),可有效平衡机体的NAD^(+)水平,改善衰老相关的病理和疾病状态.β-NMN作为抗衰老、治疗老龄性疾病的潜在活性物质,在食品和药品领域具有广阔的应用前景.与化学法相比,酶法合成β-NMN具有特异性强、高效、安全性高等优点,成为合成β-NMN的首选方法.现有酶催化合成β-NMN的路线报道不够全面,缺乏关于关键酶结构特征和催化特性等方面的研究报道.鉴于此,综述了β-NMN的4个酶催化合成路线及其关键酶的结构特征和催化特性,分析了不同酶法合成的优缺点及工业化应用的可行性.最后,讨论了现阶段酶催化合成β-NMN面临的难点和未来发展方向,以期为β-NMN的工艺优化和生产应用提供借鉴.NAD^(+)in mammals decreases with age at a systemic level,leading to the development of many age-related physiological function decline and diseases.Supplement ofβ-NMN,the intermediate of NAD^(+),can effectively restore NAD^(+)levels in aging bodies to improve their pathological and disease conditions.As a potential active substance of anti-aging and degenerative disease,β-NMN has a broad application prospect in the field of food and medicine.Compared to chemical method,enzymatic synthesis has the advantages of stronger specificity,higher efficiency and higher safety,so it has become the preferred method for production ofβ-NMN.But on the one hand,reports on the available catalytic synthesis routs ofβ-NMN are not comprehensive enough,on the other hand,there are few reports on the structure and catalytic mechanism of key enzymes.Therefore,in this paper four routs of enzymatic synthesis ofβ-NMN and their structure characteristics and catalytic mechanisms of key enzymes are reviewed.The advantages and disadvantages of different enzymatic catalytic methods and industrial advantages are analyzed.Finally,the challenge faced and the development direction of enzymatic synthesis ofβ-NMN are discussed,with anattempt to provide a reference for the process optimization and production application ofβ-NMN.
