摘  要:竹红菌甲素是重要且稀缺的药用成分,为了解丽水产竹红菌素野生竹黄菌(Shiraia bambusicola)的生物学特性,采集浙江丽水两地野生菌株子实体,分别发酵培养两地菌株QJ-17和JT-21,测定其生物量、竹红菌甲素量并克隆分析合成竹红菌甲素的O-甲基转移酶基因(Omef)序列及相对表达量。结果表明:JT-21株生物量比QJ-17株高15%、产竹红菌甲素量为QJ-17株的3.6倍,且乙醇浸提JT-21和QJ-17株竹红菌甲素量分别为丙酮的61.34%和87.75%。JT-21株Omef2和QJ-17株Omef1 CDS序列一致性达99.9%,两者蛋白质序列完全一致,但Omef2表达水平却为Omef1的3倍。这一研究阐明了丽水野生竹黄菌产竹红菌甲素量及其合成基因Omef表达水平存在差异,为丽水高产竹红菌甲素的野生竹黄菌筛选提供了依据。Hypocrellin A is a fundamental medicinal component with extremely limited source.In order to figure down the hypocrellin A production traits in wild Shiraia bambusicola strains in Lishui,Zhejiang,wild strain fruiting bodies were collected.Biomass and hypocrellin A assays were performed on two strains QJ-17 and JT-21 isolated from two villages respectively after fractional and fermentative cultivation.O-methyltransferase(Omef)gene involved in hypocrellin A synthesis was cloned and the sequences and expression level were analyzed.The collected mycelia biomass of JT-21 was 15%higher than that of QJ-17;and the quantity of leached hypocrellin A of JT-21 was 3.6 times that of QJ-17.Moreover,the amount of hypocrellin A leached with ethanol from JT-21 or QJ-17 was 61.34%and 87.75%of that leached with acetone,respectively.The CDSs of Omef2(JT-21)and Omef1(QJ-17)had 99.9%sequence identity and the protein sequences of Omef1 and Omef2 were totally identical.However,the relative expression level of Omef2 was 3 times as much as that of Omef1.This study elucidated the quantity of hypocrellin A and expression level of Omef involved in hypocrellin A synthesis were varied among wild strains in Lishui,providing a foundation for screening strains with high hypocrellin A production.
