摘 要:为了阐明绵羊卵泡颗粒细胞功能调控机制,试验选取不同条件培养的绵羊卵泡壁层颗粒细胞(MGCs)和卵丘颗粒细胞(CCs),分别设置对照组、FSH组和GDF9+FSH组,利用qPCR、Western blot和免疫荧光法分别检测MGCs和CCs中NPPC和NPR2的mRNA和蛋白表达量。结果表明:对于壁层颗粒细胞,FSH组NPPC 12 h mRNA及12,24 h蛋白水平显著高于对照组(P<0.05),极显著低于GDF9+FSH组(P<0.01);体外培养16 h后,与对照组相比,FSH组NPR2 mRNA及蛋白表达水平显著提高(P<0.05),但与GDF9+FSH组差异不显著(P>0.05)。对于卵丘颗粒细胞,FSH组NPPC mRNA及蛋白水平极显著高于对照组和GDF9+FSH组(P<0.01);FSH组NPR2 mRNA及蛋白表达水平显著高于对照组和GDF9+FSH组(P<0.05)。说明FSH可以显著提高体外培养的绵羊卵泡颗粒细胞中NPPC、NPR2的表达;GDF9可以促进FSH诱导的壁层颗粒细胞中NPPC的表达,但抑制了FSH诱导的卵丘细胞颗粒细胞中NPR2的表达。In order to provide a basis for clarifying the functional regulation mechanism of sheep follicular granulosa cells,sheep mural granulosa cells(MGCs)and cumulus cells(CCs)cultured under different conditions were selected,and control group,FSH group and GDF9+FSH group were set respectively.The mRNA and protein expressions of NPPC and NPR2 in MGCs and CCs were detected by qPCR,Western blot and immunofluorescence,respectively.The results showed that the 12 h mRNA and 12,24 h protein levels of NPPC in MGCs in FSH group were significantly higher than those in control group(P<0.05),extremly lower than those in GDF9+FSH group(P<0.01).After 16 h in vitro culture,FSH significantly increased the mRNA and protein expression levels of NPR2 in MGCs compared with the control group(P<0.05),but the differences were not significant compared with GDF9+FSH group(P>0.05).The mRNA and protein levels of NPPC in CCs of FSH group were extremly higher than those in control group and GDF9+FSH group(P<0.01).The mRNA and protein expression levels of NPR2 in FSH group were significantly higher than those in control group and GDF9+FSH group(P<0.05).These results indicated that FSH could significantly increase the expressions of NPPC and NPR2 in sheep follicular granulosa cells cultured in vitro.GDF9 promoted the expression of NPPC in FSH-induced parietal MGCs,but inhibited the expression of NPR2 in FSH-induced CCs.
