摘  要:为了解已经开展禽白血病净化的广西地方鸡种瑶鸡中禽白血病病毒(Avian leukosis virus,ALV)与禽网状内皮组织增殖病病毒(Reticuloendotheliosis virus,REV)的感染情况,试验对鸡血浆样品接种的细胞培养物分别进行ALV群特异性抗原P27的ELISA检测和亚群特异性PCR鉴定,以及REV PCR鉴定。结果显示:一份样品为ALV-J和REV的混合感染,其中ALV-J命名为GX20YLJ1,REV命名为GX20YLR1;遗传进化分析显示,分离株GX20YLJ1和GX20YLR1分别属于clade 1.3的ALV-J和罕见的Ⅱ型REV;GX20YLJ1 gp37和GX20YLR1gp20亚单位的二级结构功能区由α-Helix组成,而GX20YLJ1 gp85和GX20YLR1 gp90亚单位的二级结构具有多样性;分离株GX20YLR1的gp90与美国REVⅡ型分离株SNV的gp90亚单位的二级结构最相似,与核苷酸序列分析的进化树结果一致。结果表明,广西地方品种净化鸡群仍存在clade 1.3的ALV-J,并首次发现与Ⅱ型REV的混合感染,提示在净化检测过程中要重视与REV等其他病原混合感染的检测,同时从基因及蛋白结构层的分析应更加关注gp85和gp90两个亚单位的遗传进化。In order to understand the infection of avian leukosis virus(ALV) and reticuloendotheliosis virus(REV) in Guangxi local chicken breed Yao-chicken that has been carried out the eradication program, a series of laboratory tests were conducted, including ELISA detection of ALV group specific antigen P27 in DF-1 cell culture supernatants inoculated with chicken plasma samples and identification of ALV by subgroup-specific PCR and of REV by PCR. The results showed that a sample of ALV-J and REV co-infection was identified, and the isolates were named as GX20YLJ1 and GX20YLR1, respectively. The genetic evolution analysis showed that GX20YLJ1 and GX20YLR1 belonged to clade 1.3 ALV-J and the rare typeⅡ REV, respectively. AlphaFlod2 software predicted that the secondary structure functional regions of GX20YLJ1 gp37 and GX20YLR1 gp20subunits were composed of α-Helix, while those of GX20YLJ1 gp85 and GX20YLR1 gp90 subunits were diverse. The secondary structure of gp90 of GX20YLR1 was most similar to that of SNV that belongs to USA type Ⅱ REV, which was consistent with the results of the analysis by the sequence evolution tree. The results demonstrated that the continued existence of clade 1.3 ALV-J in Guangxi local chicken breed that has been carried out the eradication program, and its co-infection with type Ⅱ REV was found for the first time, suggesting that more attention should be paid to the detection of co-infection of ALV-J with REV and other pathogens in the process of eradication. At the same time, more attention should be paid to the genetic evolution of gp85 and gp90 subunits of the isolates at the levels of gene and protein structure.
