摘  要:【目的】利用楸树间接器官发生途径培养愈伤组织并诱导出芽是楸树良种繁育的高效技术方法,同时也为楸树遗传转化方面的研究提供技术参考。【方法】以楸树试管苗叶柄为外植体,采用L9(43)正交试验设计分别研究了不同基本培养基、植物生长调节剂浓度和光质等对愈伤组织诱导的影响,并对愈伤组织进行形态学和组织细胞学观察,据此筛选出适宜的诱导培养基,继而探索愈伤组织再分化的培养条件。【结果】在叶柄愈伤组织诱导过程中,基本培养基种类(MS、N6、DKW)的影响效应不显著,而6-BA浓度、NAA浓度和光质对愈伤组织的诱导均具有显著影响。极差分析结果显示,当培养条件为白光,植物生长调节剂6-BA与NAA的浓度比在10∶1时最有利于愈伤组织的诱导,即楸树叶柄诱导愈伤组织的最佳培养基为MS+1.0 mg·L^(-1)6-BA+0.10 mg·L^(-1) NAA,光质为白光;对9组愈伤组织制作石蜡切片,显微观察发现蓝光照射条件下,采用DKW+3.0 mg·L^(-1)6-BA+0.01 mg·L^(-1) NAA配方诱导出的愈伤组织细胞具有明显的拟分生组织,此愈伤组织在DKW+0.2 mg·L^(-1) TDZ+0.01 mg·L^(-1) NAA配方下可以成功诱导出不定芽。【结论】1)光照条件在愈伤组织诱导过程中具有显著的影响;2)TDZ具有较高的活性,能够高效地促进楸树愈伤组织诱导不定芽生长。本研究初步建立了楸树叶柄愈伤组织诱导方法和不定芽诱导的有效培养条件,为楸树无菌苗培育及遗传转化体系建立提供技术支撑。【Objective】The use of the indirect organogenesis pathway of Catalpa bungei C.A.Mey in the cultivation of callus and the induction of buds is an efficient technical method for the breeding of Catalpa bungei C.A.Mey,which provides a technical reference for the research on genetic transformation of Catalpa bungei C.A.Mey.【Method】The effects of different basic media,plant growth regulator concentrations and light quality on the induction of callus were investigated by using Catalpa bungei C.A.Mey seedling petioles as the explants in an L9(43)orthogonal test design,and the morphological and histocytological observation of the induced callus was carried out to screen for the suitable induction media and re-differentiation conditions for callus.【Result】The effect of the basic medium(MS,N6,DKW)was not significant,while the concentration of 6-BA,NAA and light quality had significant effects during the induction of petiole callus.The range analysis showed that when the cultivation conditions were white light and a concentration ratio of plant growth regulators of 6-BA to NAA as 10∶1,it was conducive to the induction of callus.That is,the best medium for the induction of callus in Catalpa bungei C.A.Mey petioles was MS+1.0 mg·L^(-1)6-BA+0.10 mg·L^(-1) NAA,and the light condition was white light.The paraffin sections were prepared for 9 groups of callus,and the microscopic observation revealed that the callus cells induced by the formulation of DKW+3.0 mg·L^(-1)6-BA+0.01 mg·L^(-1) NAA under the blue light had obvious meristemoids,and this callus could successfully produce adventitious buds under the formulation of DKW+0.2 mg·L^(-1) TDZ+0.01 mg·L^(-1) NAA.【Conclusion】1)The light condition has a significant effect on callus induction.2)TDZ has a high activity and can efficiently promote the induction of adventitious buds.In this study,we initially established a method for the induction of petiole callus and effective cultivation conditions for the induction of adventitious buds of Catalpa bungei C.A.Mey
