摘  要:目的建立骨质疏松湿寒病证动物模型,探讨湿寒证对绝经后骨质疏松症模型大鼠下丘脑弓状核β-EP的影响。方法将30只雌性SD大鼠随机分为5组,每组6只。正常对照组常规饲养,假手术组做假手术,湿寒证组加湿寒证病因因素(湿寒环境+寒凉性饲料+冷水游泳),去卵巢(OVX)模型组做OVX手术,OVX+湿寒证组做OVX手术加湿寒证病因因素。观察各组大鼠外在表征,每两天测大鼠体重、饮食量变化。9周后取材,Micro-CT检测骨密度和微结构,HE染色观察下丘脑神经细胞病理改变,免疫组化测定β-内啡肽(β-endorphin,β-EP)阳性表达水平。结果与正常对照组比湿寒证模型组大鼠出现体形臃肿,小便色清,大便塘稀等湿寒证表现;与OVX模型组相比,OVX+湿寒证模型组骨密度下降(P>0.05),但骨小梁分离度上升、骨小梁分离度标准差、结构模型指数上升,差异有统计学意义(P<0.05),表明骨微结构有改变;与正常对照组比,湿寒证模型组β-EP免疫反应阳性表达降低(P<0.05);与OVX模型组比,OVX+湿寒证模型组β-EP免疫反应阳性表达降低(P<0.05)。结论湿寒证可引起绝经后大鼠骨密度减少、骨形态计量学指标改变,可能是通过神经元细胞形态结构改变及β-EP表达水平下降间接诱发骨质疏松症的发生发展。Objective To establish an animal model of dampness-cold syndrome of osteoporosis,and to investigate the effect of dampness-cold syndrome onβ-EP in hypothalamic arcuate nucleus of postmenopausal osteoporosis model rats.Methods 30 female SD rats were randomly divided into 5 groups with 6 rats in each group.The normal control group was fed conventionally;the sham operation group was given sham operation;the dampness-cold syndrome group was given etiological factors of dampness-cold syndrome(dampness-cold environment+cold feed+cold water swimming);the ovariectomized(OVX)model group was given OVX operation;and the OVX+dampness-cold syndrome group was given etiological factors of dampness-cold syndrome.The external signs of the rats were observed,and the changes of body weight and diet were measured every 2 days.After 9 weeks,bone density and microstructure were detected by Micro-CT.Pathological changes of hypothalamic nerve cells were observed by HE staining.Positive expression ofβ-endorphin(β-EP)was determined by immunohistochemistry.Results Compared with normal control group,the rats in model group with dampness-cold syndrome showed bloatness,clear urine color and loose stool.Compared with OVX model group,bone mineral density was decreased in OVX+damp cold model group(P>0.05),but bone trabecular separation degree,standard deviation of bone trabecular separation degree and structural model index was increased,with statistical significance(P<0.05),indicating changes in bone microstructure.Compared with the normal control group,theβ-EP positive expression in model group with dampness-cold syndrome was decreased(P<0.05);Compared with OVX model group,β-EP positive expression in OVX+damp-cold syndrome model group was decreased(P<0.05).Conclusion The dampness-cold syndrome can cause the decrease of bone mineral density and the change of bone morphometric index in postmenopausal rats,which may indirectly induce the occurrence and development of osteoporosis through the change of morphological structure of neuron cells
