摘  要:【目的】为了验证脱落酸候选受体FaPYL2在草莓果实成熟过程中的作用。【方法】以八倍体‘蒙特瑞’草莓(Fragaria×ananassa cv.‘Monterey’)果实作为研究对象,利用农杆菌介导法瞬时果实侵染、亚细胞定位及等温滴定量热仪试验对FaPYL2进行功能鉴定。【结果】FaPYL2基因的表达随着草莓果实成熟而增加,并在片红期达到峰值。过表达FaPYL2基因促进果实成熟,增加花色苷和可溶性固形物积累,降低果实硬度,而沉默FaPYL2基因结果相反。另外,定位于细胞质中的FaPYL2直接与脱落酸结合,解离常数为67.56μmol/L,符合受体结合特性。【结论】FaPYL2作为一个潜在的脱落酸受体,正调控草莓果实的成熟。[Objective]This experiment is to verify the role of candidate receptor FaPYL2 of abscisic acid in strawberry fruit ripening [Methods]The present study is used the octoploid strawberry(Fragaria × ananassa cv. ‘Monterey’) fruit for studying the functional traits of FaPYL2 by Agrobacterium-mediated transient fruit infestation, subcellular localization, and isothermal titration calorimetry(ITC) experiments.[Results]FaPYL2 expression was increased with the ripening of strawberry fruit, and reached the peak at the partial red(PR) stage. Overexpression of FaPYL2 promoted fruit ripening, enhanced anthocyanins and soluble solids accumulation, meanwhile decreased fruit firmness;while FaPYL2 gene silencing through RNAi led to the opposite results. In addition, FaPYL2 is localized in the cytoplasm and binds to ABA with a Kd value of 67.56 μmol/L, consistent with receptor binding properties. [Conclusion]FaPYL2 is a potential ABA receptor that positively regulates strawberry fruit ripening.
