摘 要:目的:对阴道内主要优势乳杆菌以及阴道常见致病菌进行体外苦参总碱干预实验,观察其对细菌体外生长影响,明确苦参膜对阴道微生态恢复作用。方法:通过2,3,5-三苯基氯化四氮唑(TTC)染色法测定不同浓度的苦参膜主要有效成分苦参总碱对阴道常见致病菌金黄色葡萄球菌、大肠埃希氏、白色念珠菌和阴道常见优势乳杆菌卷曲乳杆菌、加氏乳杆菌、鼠李糖乳杆菌、罗伊氏乳杆菌的最低抑菌浓度(MIC);牛津杯法测定对致病菌抑菌活性;直径法测定优势菌菌落生长直径变化。结果:苦参总碱对金黄色葡萄球菌MIC为12.5 mg/mL,对大肠杆菌MIC为25 mg/mL,对白色念珠菌MIC为50 mg/mL,对卷曲乳杆菌MIC为12.5 mg/mL,对鼠李糖乳杆菌及罗伊氏乳杆菌的MIC为100 mg/mL,对加式乳杆菌的MIC为6.25 mg/mL。当苦参总碱浓度低于MIC值时,对阴道常见致病菌生长均有抑制作用,对鼠李糖乳杆菌具有促进增殖的作用,对卷曲乳杆菌、加氏乳杆菌及罗伊氏乳杆菌具有一定抑制作用。结论:临床给药剂量下,苦参膜能抑制阴道常见致病菌生长但不抑制阴道优势乳杆菌生长,可作为天然抗生素用于阴道炎治疗,并调节阴道微生态失衡。In this study we observed the effect of matrine on the growth of common vaginal bacteria in vitro, and clarified the recovery effect of matrine on vaginal microecology.2,3,5-triphenyltetrazolium chloride(TTC)staining method was used to determine the minimum inhibitory concentration(MIC)of matrine against common vaginal pathogenic bacteria such as Staphylococcus aureus,Escherichia coli,and Candida albicans,and common vaginal dominant bacteria such as Lactobacillus crispatus,Lactobacillus gasseri,Lactobacillus rhamnosus and Lactobacillus reuteri.Oxford cup method was used to determine the antibacterial activity of matrine against pathogenic bacteria.The variation of colony diameter of the dominant bacteria was determined by diameter method.The MIC of matrine was 12.5 mg/mL against Staphylococcus aureus,25 mg/mL against Escherichia coli,50 mg/mL against Candida albicans,12.5 mg/mL against Lactobacillus crispatus,100 mg/mL against Lactobacillus rhamnosus and Lactobacillus reuteri,and 6.25 mg/mL against Lactobacillus gasseri.When the concentration of matrine was lower than the MIC value, it could inhibit the growth of common vaginal pathogens, promote the proliferation of Lactobacillus rhamnosus,and inhibit Lactobacillus crispatus, Lactobacillus gasseri and Lactobacillus reuteri.In conclusion, at clinical dosage, matrine can inhibit the growth of common vaginal pathogenic bacteria, but does not inhibit the growth of dominant vaginal Lactobacillus,which can be used as a natural antibiotic for the treatment of vaginitis and vaginal microecological unbalance.
