摘　　要：目的:研究驴皮胶及常见伪品胶马皮胶和牛皮胶的蛋白凝胶电泳行为。方法:自制3种皮胶,计算出胶率,BCA蛋白浓度测定试剂盒测定其蛋白含量,计算上样量,使上样量为20~30μg。垂直电泳仪进行SDS-PAGE凝胶电泳,采用考马斯亮蓝及银染法显色。结果:3种皮胶的凝胶电泳行为相似,蛋白成分主要集中在分子量大于25 kDa段上,银染法灵敏度更高,可显示较多条带。结论:3种皮胶分子量分布宽泛,具有共同的条带,进一步可用多肽蛋白质谱鉴定技术对各混合物组分进行更深入分析。Objective:To study the protein gel electrophoretic behavior of donkey-hide glue and common fake glue such as horse-hide glue and cow-hide glue.Methods:Three kinds of hide glue were prepared,the glue rate was calculated,the protein content was determined by BCA protein concentration determination kit,and the sample loading was calculated to make the sample loading 20~30μg.SDS-PAGE gel electrophoresis was performed by vertical electrophoresis apparatus,and color was developed by Coomassie brilliant blue and silver staining.Results:The gel electrophoresis behaviors of the three kinds of hide glue were similar.The protein components were mainly concentrated on the segments with molecular weight greater than 25 kDa.The silver staining method was more sensitive and showed more bands.Conclusion:The molecular weight distribution of the three kinds of hide glue is broad,and they have common bands.The peptide protein mass spectrometry can be used to further analyze the components of each mixture.
