摘 要:目的 探讨细菌性腹膜炎大鼠黏膜炎症因子与应激状态及其Wnt5a/β-catenin信号通路的变化。方法 对大鼠建立细菌性腹膜炎模型,并将造模成功的实验性细菌性腹膜炎大鼠分为8 h、16 h、24 h模型组,每组10只,于注射菌液后8 h、16 h、24 h时检测各组大鼠体征变化、血清内毒素,观察腹腔黏膜病理变化,检测黏膜炎症因子白细胞介素-1β(IL-1β)、IL-4、IL-6、IL-10、肿瘤坏死因子-α(TNF-α)水平,与应激因子超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、活性氧自由基(ROS)水平,并检测黏膜组织中Wnt5a/β-catenin信号通路相关蛋白与mRNA水平。结果 对照组疾病活动指数(DAI)评估、内毒素水平低于模型组;对照组黏膜结构清晰,未见充血水肿、炎症浸润,模型组黏膜上皮细胞有明显坏死脱落,出现水肿、出血,大量炎性细胞浸润;模型组IL-1β、IL-4、IL-6、TNF-α表达高于对照组,IL-10低于对照组(P<0.001);模型组MDA、ROS高于对照组,SOD、GSH-Px低于对照组(P<0.001);模型组的Wnt5a/β-catenin蛋白、mRNA表达高于对照组(P<0.001)。结论 Wnt5a/β-catenin信号通路与细菌性腹膜炎大鼠的黏膜炎症与应激状态有关,其起效机制可能是Wnt5a/β-catenin激活炎症细胞释放IL-1β、IL-6、TNF-α等促炎因子,抑炎因子IL-10释放减少,进而介导黏膜出现应激损伤。OBJECTIVE To investigate the changes of mucosal inflammatory factors,stress status and their Wnt5a/β-catenin signaling pathway in rats with bacterial peritonitis.METHODS The bacterial peritonitis model was established in rats,and rats with successful experimental bacterial peritonitis were divided into 8 h,16 h and 24 h model groups,with 10 rats in each group.The changes in physical signs and serum endotoxin of the rats in each group were detected at 8 h,16 h,and 24 h after the injection of the bacterial solution,the pathological changes of the peritoneal mucosa were observed,and the expression levels of mucosal inflammatory factors IL-1β,IL-4,IL-6,IL-10 and TNF-α,and the levels of stress factors SOD,MDA,GSH-Px and ROS,as well as Wnt5a/β-catenin signaling pathway-related proteins and mRNA levels in mucosal tissue were detected.RESULTS The DAI assessment and endotoxin level in the control group were lower than those in the model group;the mucosal structure of rats in the control group was clear,and there was no congestion,edema,and inflammatory infiltration,while the mucosal epithelial cells in the model group had significant necrosis and detachment,edema,hemorrhage,and massive inflammatory cell infiltration;the expression levels of IL-1β,IL-4,IL-6 and TNF-α in the model group were higher than those in the control group,while the expression level of IL-10 was lower than that in the control group(P<0.001);the levels of MDA and ROS in the model group were higher than those in the control group,and the levels of SOD and GSH-Px were lower than those in the control group(P<0.001);the expression of Wnt5a/β-catenin protein and mRNA in the model group were higher than those in the control group(P<0.001).CONCLUSION The Wnt5a/β-catenin signaling pathway was associated with mucosal inflammation and stress state in rats with bacterial peritonitis,and its mechanism of initiation might be the activation of inflammatory cells to release IL-1β,IL-6 and TNF-α induced by Wnt5a/β-catenin,and decrease the release of anti-i
