摘  要:目的:探讨大黄素在改善骨关节炎软骨降解中的作用及机制。方法:大黄素(20µmol/L)预处理SD大鼠软骨细胞2 h,IL-1β(10 ng/ml)孵育24 h。MTT检测不同处理组细胞活力,RT-PCR和Western blot分析aggrecan、collagenⅡ、MMP-13、ADAMTS-4、NF-κB p65、IKK-β、IκB-α表达。结果:与IL-1β组相比,IL-1β+大黄素组大鼠软骨细胞aggrecan和COL2A1 mRNA水平显著升高(P<0.05),MMP-13、ADAMTS-4、NF-κB p65和IKK-βmRNA水平显著降低(P<0.05),而IκB-αmRNA水平显著升高(P<0.05)。结论:大黄素通过抑制NF-κB途径抑制MMP和ADAMTS表达,从而发挥软骨保护作用。Objective:To investigate effect and mechanism of emodin on improving cartilage degradation of osteoarthritis.Methods:SD rat chondrocytes were pretreated with emodin(20μmol/L)for 2 h and incubated by IL-1β(10 ng/ml)for 24 h.Cells viabilities of different treatment groups were detected by MTT.Expressions of aggrecan,collagenⅡ,MMP-13,ADAMTS-4,NF-κB p65,IKK-βand IκB-αwere analyzed by RT-PCR and Western blot.Results:Compared with IL-1βgroup,levels of aggrecan and COL2A1 mRNA of rat chondrocytes in IL-1β+emodin group were significantly increased(P<0.05),levels of MMP-13 and ADAMTS-4,NF-κB p65 and IKK-βmRNA were significantly reduced,while IκB-αmRNA level was significantly increased(P<0.05).Conclu⁃sion:Emodin inhibits expressions of MMP and ADAMTS by inhibition of NF-κB pathway,thereby exerting chondroprotective effect.
