摘 要:当生物体内环境偏离正常范围,或调控矿化过程的大分子无法发挥功能时,过饱和的矿物质在组织表面成核并生长,或从体液中析出并附着组织表面,引发炎症、生理组织功能损坏和血管、尿管等堵塞。医疗长期植入物如何获得生物组织甚至超越生物组织的抗污染、抗结垢能力需要被重视。以上问题的解决亟需结晶热力学和界面晶体工程的研究,具有化学工程、材料科学和医学学科交叉的特点。本论文从改变体液环境和植入材料表面改性调控结晶的思路出发,介绍了病理性矿化机制,综述了病理矿化诱发的几种重要结石疾病的结晶调控方法,重点阐述了能够抑制病理矿化的小分子、大分子、纳米颗粒和聚合物表面抑制结晶机制以及设计开发策略。最后,总结了抑制病理矿化领域的主要问题,对抑制剂分子开发和生物防垢表面设计的发展提出展望。Minerals crystallization in body fluids and adhering to the surface of human tissues,causing inflammation,urinary calculi,blockage of the vessel,and damage to many physiological function when the biological environment deviates from normal or macromolecules fail to regulate the mineralization process.Thus,it is important to improve medical implantation anti-fouling and anti-scaling ability.To solve the above problems,it is urged to improve the thermodynamic and interface engineering of crystallization,which has outstanding interdisciplinary characteristics in the fields of chemical engineering,materials science and medicine.This paper starts with two strategies:change the environment of body fluids and design the surface of medical implant materials.Mechanisms of pathological mineralization are described.Crystallization regulation methods are then reviewed.Small molecules,macromolecules,nanoparticles,and polymer surfaces capable of inhibiting pathological mineralization are highlighted,and strategies for design effective inhibitors and mechanisms of inhibition of crystallization are discussed.The main problems in the inhibiting pathological mineralization are summarized,and the prospects on the development of inhibitor molecules and the design of antifouling surface are put forward.
