摘 要:同居感染是在实验室条件下模拟自然感染的一种有效途径,为建立兔病毒性出血症病毒(RHDV)同居感染模型,取血凝价为1∶29的RHDV阳性兔的肝研磨液颈部皮下注射接种家兔,将该接种兔与未接种的健康家兔同笼饲养,并观察实验家兔的临床症状、病理解剖变化以及检测实验家兔肝组织中的病毒。注射接种RHDV组家兔约注射后24 h出现该病典型临床症状,同居感染家兔在同居后约36~48 h时陆续出现典型临床症状;注射感染组家兔在接种后约38 h死亡,同居感染家兔在同居后72~80 h陆续死亡;同居感染家兔剖检后可见肝、脾和肺出现明显的病变,其与注射感染组家兔相似,均符合RHDV感染导致家兔死亡的典型剖检变化;RT-PCR检测结果显示,注射感染和同居感染家兔的肝组织中均可检测到RHDV。研究表明,本研究成功建立了RHDV同居感染模型,为疫苗研制和病毒致病机制的研究奠定了基础。Laboratory cohabitation challenge model is an effective way to simulate natural infection.In order to establish the model for RHD caused by RHDV,the liver grinding fluid of RHDV-positive rabbits with a hemagglutination value of 1∶2~9 was used to inoculate the rabbits subcutaneously in the neck,and the inoculated rabbits were raised with uninoculated rabbits in the same cages.The clinical symptoms,pathological and anatomical changes of the experimental rabbits were observed,and the virus in the liver tissue of the experimental rabbits were detected.The results showed that the typical clinical symptoms of injected rabbits at about 24 h after inoculation,and the similar clinical symptoms appeared in cohabiting rabbits at about 36-48 h after cohabitation.Injected rabbits died about 38 h after inoculation,and cohabiting rabbits died 72-80 h after cohabitation.Significant pathological changes in liver,spleen and lung were observed both in injected rabbits and cohabitation rabbits.The results of RT-PCR showed that RHDV could be detected in the liver tissues of rabbits in two groups.Researches show that,the cohabitation infection model of RHDV was successfully established in this study,which laid a foundation for the development of vaccine and the study of the pathogenesis of the virus.
