肉鸡骨髓间充质干细胞的分离纯化及鉴定

摘　　要：通过全骨髓差速贴壁培养法分离培养肉鸡BMSC,对其进行成骨及成脂诱导分化试验,并通过茜素红S染色、ALP及成骨代谢相关基因检测成骨分化情况,油红O染色及脂代谢相关基因检测成脂分化情况。结果,BMSC在进一步纯化的过程中细胞形态由不规则形趋近并统一于成纤维状,同时其生长曲线呈“S”型,并随着代数的增加,倍增时间逐渐延长。体外诱导分化结果显示,BMSC具有多向分化潜能,可分化为成骨细胞和脂肪细胞。综上所述,本试验分离纯化的肉鸡BMSC具有易于分离、扩增及多向分化潜能的特性,为肉鸡BMSC的提取及鉴定提供了理论依据。BMSC were isolated and cultured by whole bone marrow differential apposition culture method,and osteogenic and lipogenic induction differentiation assays were performed.The osteogenic differentiation was detected by alizarin red S staining,ALP and genes related to osteogenic metabolism,and lipogenic differentiation was detected by oil red O staining and genes related to lipid metabolism.The results showed that the cell morphology of BMSC tended to be irregular and unified into a fibrous shape during further purification,while the growth curve showed an“S”shape,and the ploidy time was gradually prolonged with the increase of the number of generations.The induction of differentiation in vitro showed that the BMSC had multi-directional differentiation potential and could be differentiated into osteoblasts and adipocytes.In conclusion,the purified broiler BMSC were easy to isolate,amplify and have multi-directional differentiation potential,which provided the theoretical basis for the extraction and identification of broiler BMSC.
