摘  要:本研究以茄子单性结实品系D-10和非单性结实品系03-2所构建的抑制差减文库(SSH)中差异表达的EST片段fan11为依据,利用RACE技术克隆得到一条与AGL9基因一致性达97.93%的cDNA序列,命名为SmA GL9。生物信息学分析表明,该基因有一个全长726 bp的开放阅读框,编码241个氨基酸,预测蛋白分子量为27.546 2 kD,理论等电点为9.00。保守结构域分析发现该蛋白含有MADS-MEF2-like和K-box两个保守结构域,属MADS-box转录因子。同源进化分析结果显示SmAGL9基因的蛋白质序列与辣椒AGL9基因同源关系相近。RT-qPCR分析表明,低温条件下SmAGL9基因在单性结实和非单性结实的子房和果实发育过程中均有表达,果实发育初期单性结实品系中的表达量显著高于非单性结实品系,推测SmAGL9基因在茄子单性结实子房和果实发育初期具有重要的调控作用。结果可为进一步研究茄子单性结实果实形成的分子机理提供参考。Based on th e EST fan11 from eggplant suppression subtractive hybridization library constructed by parthenocarpic line D-10 and non-parthenocarpic line 03-2, a gene named SmA GL9, which was similar to AGL9gene with the similarity of 97.93%, was cloned by rapid amplification of cDNA ends(RACE). Bioinformatics analysis indicated that the SmA GL9 gene containing an open reading frame of 726 bp, encoding a protein with 241amino acids, and the protein molecular mass was 27.5 kD, the isoelectric pI was 9.00. SmA GL9 was a MADS-box gene, which owns the conserved domain of MADS-MEF2-like and K-box, and phylogenetic tree analyses showed that the amino acid of SmA GL9 has the highest similarity with the pepper AGL9. RT-qPCR analysis showed that the SmA GL9 gene was expressed throughout the development of eggplant fruit in D-10 and 03-2, and it as higher in parthenocarpy at the early stage of fruit development, so it was speculated that the SmA GL9 gene has a role in the development of parthenocarpy fruit in eggplant, which provides a reference for further research on the molecular mechanism of the formation of parthenocarpic eggplant.
