摘 要:果实品质一直都是番茄遗传育种研究领域的热点。为了挖掘新的相关QTLs,本研究以191份栽培番茄为试材,包括24份樱桃番茄、108份常规品种和59份现代品系,对番茄单果重、果实横径、果实形状、心室数、糖含量和硬度6个品质性状进行两个年份的表型分析。结果显示所有性状均表现出丰富的表型变异,其中单果重、果实横径、果实形状和心室数在三个群体间均差异显著。选取273个分子标记(120个SNP, 128个In Del和25个SSR)对所有试材进行基因分型,群体结构分析显示所有试材可分为3个亚群体,与预先划分的3个群体基本一致。利用基于全基因组扫描的策略对6个性状进行关联作图,共获得与目标性状相关联的分子标记33个,与硬度相关联的标记最多(8),与果实横径关联的标记最少(3)。总共有17个关联标记(51.5%)与前人报道的QTLs/基因位于相同的位置。本研究结果可以与前人发现的相关QTLs相互验证,同时也检测到一些新的影响番茄品质的QTL位点。Fruit quality has always been a hot topic in tomato genetics and breeding. In order to explore new related QTLs, we studied a group of 191 tomato cultivars including 24 cherry tomato, 108 traditional varieties, and 59contemporary lines. Phenotypic analysis was carried out on six fruit quality traits including fruit weight, fruit diameter, fruit shape, locule number, sugar content and fruit firmness in two years. The results showed that all the traits exhibited a large range of phenotypic variation. There were significant differences in fruit weight, fruit diameter,fruit shape, and locule number among the three groups. We used 273 molecular markers, including 120 SNPs, 128InDels, and 25 SSRs, to genotype all 191 tomato accessions. Population structure analysis showed that all samples could be divided into three subpopulations corresponding to the predefined groups. Genome-wide association mapping detected 33 associated markers for six traits, fruit firmness had the largest number of associated markers(8),while fruit diameter had the fewest associated markers(3). A total of 17 associations(51.5%) were co-localized with previously-identified QTLs or known genes. The results of our study confirm some previous QTLs and identified some new loci that affect fruit quality in tomato.
