摘  要:为探究番茄斑萎病毒(tomato spotted wilt virus, TSWV)侵染感病辣椒品种‘萧新19’后病毒积累量变化规律,本研究将TSWV人工摩擦接种于‘萧新19’植株,通过症状学观察、血清学及半定量RT-PCR三者结合的方式监测接种后第1、7、14、21、28、35天(days post inoculation, dpi)的‘萧新19’新叶的发病情况。利用实时荧光定量PCR技术对6个时期样品的病毒积累量进行定量检测分析。症状学观察发现在14 dpi‘萧新19’才开始表现症状,随后病症逐渐明显,伴随有同心环纹、坏死斑点、褪绿等症状,随时间推移病症逐渐加重。血清学及半定量RT-PCR检测均表明TSWV接种成功。RT-qPCR病毒积累量的检测结果显示,在14 dpi‘萧新19’叶片病毒的积累量达到最高,在1~14 dpi里辣椒叶片病毒积累量呈上升趋势,14 dpi后病毒积累量呈下降趋势。研究表明只有当‘萧新19’新叶的TSWV量积累到一定水平时,寄主才会显症,这为下一步辣椒的早期诊断和防控提供了新思路。同时感病辣椒品种‘萧新19’的TSWV积累量整体呈现“先增后降”的变化规律,也为后续辣椒TSWV感病机制相关研究提供了理论基础。In order t o explore the variation of virus accumulation of susceptible pepper variety ’Xiaoxin19’infected by tomato spotted wilt virus(TSWV), inoculate TSWV into ’Xiaoxin19’ plants by artificial friction, and through symptomatic observation, serology and combined with the method of semi-quantitative RT-PCR to monitor the incidence of ’Xiaoxin19’ new leaves at 1, 7, 14, 21, 28, 35 dpi. The real-time fluorescent quantitative PCR technology was used to quantitatively detect and analyze the virus accumulation of samples in six periods. Symptomology observation revealed that the symptoms of ’Xiaoxin19 ’ began to appear at 14 dpi, and then the symptoms gradually became obvious, accompanied by concentric rings, necrotic spots, chlorosis and other symptoms,which gradually aggravated over time;both serology and semi-quantitative RT-PCR tests showed that TSWV was successfully inoculated;the RT-qPCR virus accumulation test results showed that the virus accumulation in’Xiaoxin19’ leaves reached the highest at 14 dpi, and the virus accumulation in pepper leaves showed an upward trend from 1 dpi to 14 dpi, and the virus accumulation showed a downward trend after 14 dpi. Studies have shown that only when the amount of TSWV in the new leaves of ’Xiaoxin19’ accumulates to a certain level, the host will be symptomatic, providing a new idea for the next step of early diagnosis and prevention of pepper. At the same time, the TSWV accumulation of the susceptible pepper variety ’Xiaoxin19 ’ showed a "increasing first and then decreasing" change pattern as a whole, which also provided a theoretical basis for subsequent research on the mechanism of pepper TSWV.
