摘 要:研究不同因子对地被菊‘珍粉’品种再生体系建立的影响,为地被菊的推广种植打下技术基础。以地被菊‘珍粉’二年生茎段为试材,研究不同消毒时间梯度下2%NaClO和0.1%HgCl_(2)对茎段消毒效果、6-BA:NAA和6-BA:KT激素组合对不定芽诱导、不同浓度IBA和NAA对不定根发生,半开瓶和全开瓶两种炼苗方式对移栽成活率的影响。结果显示,2%NaClO消毒效果优于0.1%HgCl_(2),且在6 min时萌发率最高,达到90.00%;外植体腋芽诱导率在6-BA 0.1 mg/L+KT 0.1 mg/L组合最高,达到97.67%,出芽指数为6.33,优于6-BA:NAA组合;不同浓度IBA诱导不定根发生率均达100%,高于NAA,但在根系质量和后期成活率上,IBA浓度为0.5 mg/L时最优;试管苗在半开盖炼苗5 d后的移栽成活率达到了100%,且植株长势健壮。Different factors on the establishment of the regeneration system of Chrysanthemum morifolium ’Zhenfen’ were studied to lay a technical foundation for the poplarization and cultivation in tropical regions. Two years old stem section of ’Zhenfen’ C. morifolium were used as the experimental materials. The effects of 2% NaClO and0.1% HgCl_(2) under different time gradient, induction of adventitious buds under different 6-BA:NAA and 6-BA:KT concentration combinations, the occurrence of adventitious roots under different concentrations of IBA and NAA,and the rates of survival by different modes of acclimatization and transplant were studied. The results showed that the disinfection effect of 2% NaClO was better than 0.1% HgCl_(2), and the germination rate was the highest at 6 min,reaching 90.00%. The axillary bud induction rate was the highest in 6-BA 0.1 mg/L+ KT 0.1 mg/L combination,reaching 97.67%, and the proliferation coefficient was 6.33, which was better than that in 6-BA:NAA combination. The rates of adventitious root under different concentrations of IBA all reached 100%, which was higher than that of NAA, the root quality and survival rate achieve the optimal when IBA was 0.5 mg/L meanwhile. After 5 days of transplanting, the survival rate of test tube seedlings reached 100%, and the plants grew well.
