美女樱种子诱导芽组织培养

摘　　要：以美女樱(Verbena hybrida)种子为外植体,利用组培方法诱导成苗,成苗后进行继代和生根,建立一套美女樱种子的组培体系。结果表明:试验前一晚用蒸馏水浸泡种子达到的效果较好,并且种子需要切除两端0.01 cm处以破坏种皮才可诱导成苗,若不破坏种皮便不可生苗;所有灭菌时间只有空白对照全部污染,其余的污染率均为0,当灭菌时间为17 min时成活率和生长情况最好,为61.11%;MS+6-BA 1.5 mg/L+NAA0.05 mg/L+GA30.5 mg/L培养基为最适诱导的培养基;MS+6-BA 2.0 mg/L+NAA 0.05 mg/L+GA30.5 mg/L培养基为最适继代的培养基;再生苗移植到1/2MS+NAA 0.05 mg/L培养基上,植株健壮,生根率为100%;将生根培养的美女樱幼苗移栽到营养土∶珍珠岩∶蛭石=4∶2∶1的混合基质中,成活率为82.35%。本试验研究的美女樱苗具有优良性状,既解决了内蒙古园林花卉种类单一的问题,又在运输方面大大降低成本及亏损率。本研究为将非乡土树种的美女樱快速引种到内蒙古城市园林建设中提供一定借鉴作用。Taking the seeds of Verbena hybrida as explants, the buds were induced by tissue culture, and then subculture and rooting were carried out after seedling formation. A set of tissue culture system was established for the seeds of Verbena hybrida. The results showed that: the effect of soaking seeds in distilled water the night before the experiment was better, and the seeds needed to be cut off 0.01 cm at both ends and damaged the seed coat to induce seedlings. If the seed coat was not damaged, the seedlings could not be born. All sterilization time was only the blank control, and the rest sterilization time was not polluted. When the sterilization time was 17 min, the survival rate and growth condition were the best, which was 61.11%. The results showed that MS+6-BA 1.5 mg/L+NAA 0.05 mg/L+GA30.5 mg/L was the most suitable medium for induction;MS+6-BA 2.0 mg/L+NAA 0.05 mg/L+GA30.5 mg/L was the most suitable medium for subculture;regenerated plantlets were transplanted to 1/2MS +NAA 0.05 mg/L on the medium, the plants were robust and the rooting rate was 100%. The seedlings of Verbena hybrida were transplanted into the mixed medium of nutrient soil: perlite: vermiculite=4∶2∶1, the survival rate was82.35%. The Verbena hybrida seedlings studied in this experiment have excellent characters, which not only solve the problem of single species of garden flowers in Inner Mongolia, but also greatly reduce the cost and loss rate in transportation. This study will provide a reference for the rapid introduction of non-native cherry trees to urban landscape construction in Inner Mongolia.
