摘　　要：铁线莲具有极高的观赏价值,是近年来最流行的花园植物之一,通过植物组织培养技术是实现该属植物无性繁殖和产业化生产的最佳途径。本研究以3年生的‘粉红欣喜’和‘灵感’铁线莲的带芽茎段为外植体,进行组培快繁的研究。结果表明,适于‘粉红欣喜’的初代培养基为MS+1 mg/L BA,腋芽萌发率为100%且新生枝条形态正常;增殖最佳培养基为MS+0.5 mg/L BA+0.02 mg/L NAA,增殖系数为4.54;生根最佳培养基为1/2MS+1.5 mg/L IBA+1 mg/L IAA,生根率高达76.92%。适于‘灵感’初代培养基为MS+1.5 mg/L BA,新生枝条形态正常且萌发率为100%;增殖最佳培养基为MS+1.5 mg/L BA+1 mg/L KT+0.2 mg/L NAA,增殖系数为5.76;生根最佳培养基为1/2MS+1.5 mg/L IBA+0.5 mg/L IAA,生根率高达92.31%。本研究建立了‘粉红颀喜’和‘灵感’铁线莲完整的组织培养技术体系,为铁线莲优良种质资源的推广应用提供了理论基础。Clematis is on e of the most popular garden plants with high ornamental value in recent years. Plant tissue culture technology was the best way to achieve asexual propagation and industrial production of Clematis. In order to study the tissue culture and rapid propagation of Clematis ’Pink Delight’ and INSPIRATION ’Zoin’,3-year-old stem segments with axillary buds were used as explants. The results showed that MS+1 mg/L BA was suitable for primary culture of ’Pink Delight’, axillary bud germination rate was 100% and new branch morphology was normal;the best proliferation medium was MS+0.5 mg/L BA+0.02 mg/L NAA, in which proliferation coefficient was 4.54;the best rooting medium was 1/2MS+1.5 mg/L IBA+1 mg/L IAA, rooting rate was 76.92%. And MS+1.5 mg/L BA was suitable for primary culture of INSPIRATION ’Zoin’, the morphology of new branches was normal and the germination rate was 100%;the best proliferation medium was MS+1.5 mg/L BA+1 mg/L KT+0.2 mg/L NAA, the proliferation coefficient was 5.76;the best rooting medium was 1/2MS+1.5 mg/L IBA+0.5 mg/L IAA, the rooting rate was 92.31%. This study established a complete tissue culture system of ’Pink Delight’ and INSPIRATION ’Zoin’, which provided a theoretical basis for the promotion and application of excellent germplasm resources of Clematis.
