摘 要:研究了黄连、杜仲、五倍子、白头翁4种中草药提取的最佳工艺.以乙醇为溶剂,通过超声波提取和牛津杯法,以单因素试验考察提取时间、提取温度、乙醇浓度、料液质量浓度及提取溶剂pH等条件,对大肠杆菌、枯草芽孢杆菌、金黄色葡萄球菌、酵母菌、黑曲霉的抑菌效果;并以二倍稀释法确定其最小抑菌浓度(MIC)和最小杀菌浓度(MBC).结果表明:在本实验条件下,4种中草药的最佳提取条件为:提取时间10 min,提取温度50℃,乙醇浓度60%~70%,料液质量浓度125 g/L,提取时pH为2.0.4种中草药对5种微生物的MIC值在7.8~125 mg/mL,MBC值在31.25~250 mg/mL.本研究得出的4种中草药的最佳提取工艺,为中草药饲料添加剂的科学配伍、配方研发以及标准化流程的建立奠定了理论基础.Put under study was the optimum extraction process of four Chinese herbal medicines:Coptis chinensis,Eucommia ulmoides,Chinese gallnut and Pulsatilla chinensis.With ethanol as solvent,ultrasonic extraction and Oxford cup method were used to investigate the antibacterial effects of extraction time,extraction temperature,ethanol concentration,solid-liquid mass concentration and extraction pH on Escherichia coli,Bacillus subtilis,Staphylococcus aureus,yeast and Aspergillus niger through single factor test.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)were determined by the double dilution method.Under the said experimental conditions,as the results indicated,the optimum extraction conditions of four Chinese herbal medicines were:extraction time:10 min,extraction temperature:50℃,ethanol concentration:60%~70%,solid-liquid mass concentration:125 g/L,and extraction pH 2.0.The four kinds of Chinese herbal medicine on five microbes revealed a MIC value of about 7.8~125 mg/mL,and a MBC value of about 31.25~250 mg/mL.This study obtained the best extraction process of four kinds of Chinese herbal medicine,laying a theoretical foundation for the scientific compatibility of Chinese herbal feed additives,and formulation R&D as well as the establishment of standardized process.
