摘 要:以抗寒性不同的两个冬小麦品种“东农冬麦1号”和“济麦22”为试验材料,以包含生长点的分蘖节为研究对象,分析低温驯化、封冻初期及深度封冻期两份材料细胞膜受损差异,检测分蘖节处细胞活力,对两个品种分蘖节进行蛋白质双向电泳,筛选差异表达蛋白后作质谱分析。结果表明,低温下东农冬麦1号细胞膜受损程度小于济麦22,分蘖节处细胞活力显著高于济麦22。共鉴定出品种间21个差异蛋白点,低温驯化期济麦22参与能量代谢、蛋白合成、光呼吸及抗性蛋白表达量较高,东农冬麦1号参与糖代谢、细胞壁合成的蛋白表达量高;封冻后东农冬麦1号抗性蛋白、参与能量代谢、物质合成及维持细胞稳定的蛋白表达量增强。研究明确抗寒品种东农冬麦1号耐低温蛋白表达情况,丰富寒地冬小麦安全越冬理论基础,为采用分子手段开展抗寒性遗传改良提供理论依据。Two winter wheat varieties Dongnong Dongmai 1 and Jimai 22 with different cold resistance were used as materials, and the tiller nodes including growing points were taken as the research object. The differences of cell membrane damage in the two materials during low temperature acclimation, early freezing stage and deep freezing period were analyzed, and the cell viability including tiller nodes was detected, and protein two-dimensional electrophoresis was performed on the tiller nodes of the two varieties. The differentially expressed proteins were screened and analyzed by mass spectrometry. The results showed that the damage degree of cell membrane of Dongnong Dongmai 1was less than that of Jimai 22 at low temperature, and the cell activity at tiller node was significantly stronger than that of Jimai 22. A total of 21 differential protein spots were identified. During low temperature acclimation, Jimai 22 was involved in energy metabolism, protein synthesis, photorespiration and resistance protein expression, while Dongnong Dongmai 1 was involved in sugar metabolism and cell wall synthesis. Dongnong Dongmai 1 increased the expression of resistance protein, energy metabolism, substance synthesis and cell stability after freezing. The purpose of this study was to clarify the protein expression of low temperature tolerance of cold-resistant variety Dongnong Dongmai 1,enrich the theoretical basis of safe overwintering of winter wheat in cold regions, and provide theoretical basis for genetic improvement of cold resistance by molecular means.
